摘 要:目的观察蠲痹汤对胶原诱导性关节炎(CIA)大鼠血清及滑膜组织中Fas/FasL系统的影响。方法用造模成功的牛Ⅱ型胶原诱导性关节炎大鼠14只按随机数字表法分为药物组(7只)和模型组(7只),未造模的7只作为对照组。对照组、模型组按10mL/kg灌服0.9%氯化钠注射液,药物组按38.61mg/kg灌服蠲痹汤,每日1次,连续4周。末次灌胃后取材,采用酶联免疫吸附试验(ELISA)法检测各组大鼠血清中Fas、FasL的浓度,免疫组织化学法检测滑膜组织中Fas、FasL的表达情况。结果与对照组相比,模型组和药物组血清和滑膜组织中Fas表达明显升高,FasL表达降低。与模型组相比,药物组血清和滑膜组织中Fas表达降低,FasL表达升高。结论 FasL在药物组血清和滑膜组织中表达高于模型组,而Fas表达降低,提示蠲痹汤能调节Fas/FasL系统介导的细胞凋亡机制、减缓CIA大鼠的症状,为临床治疗类风湿关节炎提供一定的理论基础。Objective To observe the effect of Juanbitang decoction on Fas/FasL system in serum and synovial tissue of collagen-induced arthritis(CIA) rats. Methods The rat model of CIA was established by injecting bovine typeⅡ collagen. Fourteen model rats were randomly divided into model group(n= 7,saline 10 mL · kg-1 · d-1 by gavage) and Juanbitang group( n =7, 38. 61 mL · kg-1 · d-1 by gavage);Seven nomal rats were included in control group (saline 10 mL · kg-1 · d-1 by gavage). All rats were garaged once per day for 4 weeks. The levels of Fas and FasL in serum were determined by ELISA and the expression of Fas and FasL in the synovial tissue was detected by immunohistochemical method after 4 weeks of intragastric administration. Results Compared with the control group, the expression of Fas was significantly increased in the serum and synovial tissue of the model group and the Juanbitang group, and FasL expression was decreased. Compared with the model group, the expression of Fas in serum and synovial tissue decreased, FasL expression increased. Conclmion The expression of FasL in serum and synovial tissue of the Juanbitang group is higher than that of the model group, and the expression of Fas is decreased, suggesting that Juanhitang decoction can regulate apoptosis mechanism mediated by Fas/FasL system, slow down the symptoms of CIA rats,which can provide a theoretical basis to treat rheumatoid arthritis.
