摘  要:目的:本研究通过观察枸杞多糖对UVB照射的体外培养人皮肤成纤维细胞活力的影响,对比探讨枸杞多糖抗光老化的作用效果。方法:以30μw/cm^2的UVA照射成纤维细胞,MTT法测定不同浓度的枸杞多糖对人纤维细胞的增殖影响,western-blot检测MMP-1、MMP-3、MMP-9的表达。结果:(1)MTT检测细胞增殖情况:在相同UVA照射条件下,模型组OD值显著降低(P<0.01),各给药组随枸杞多糖浓度的升高而增高,且高剂量组与低、中剂量组比较,差异有统计学意义(P<0.05,P<0.01);(2)免疫印迹法检测MMPs蛋白的表达结果显示,模型组MMPs蛋白表达增加,而给药组MMPs的表达明显减弱,且高剂量组显著低于中剂量组(P<0.05,P<0.01)。结论:枸杞多糖能明显增强紫外照射引起的皮肤成纤维细胞的活力,同时下调细胞中基质金属蛋白MMP-1、MMP-3、MMP-9的表达。Objective: This study was to investigate the effect expression of MMP-1, MMP-3, MMP-9 by Lycium Barbarum Polysaccharides (FBP) after UVA and UVB irradiation in vitro of fibroblasts and explore the mechanism of it. Methods: ①MTT method to detect the cells proliferation activity by 30mJ/cm^2 UVA irradiation; ②Westem-blot method to detect the expression of MMP-9 protein, MMP-3, of theMMP-1 of cells after FBP acted on. Results: ①MTT method showed that, compared with the blank group, the OD value of the model group significantly decreased (P〈 0. 01 ), OD values in each group and the blank group was significantly decreased (P 〈 0.05 ) ; compared with the model group, besides the technical FBP low dose group, the OD value of the rest groups were significantly elevated; ②The expression of human fibroblast cell MMPs protein by Western blot results showed that, compared with the blank group, expression of MMPs protein of the model group increased, and expression of the drug groups decreased signifi- cantly, and the high dose of FBP was better than that of the low dose of FBP. Conclusion: The polysaccharide could obviously increase the activity of skin fibroblasts induced by ultraviolet irradiation, and down regulate the expression of MMP-9, MMP-3 and MMP-1.
