摘  要:目的观察麝香保心丸改善心肌梗死小鼠外周微循环障碍的作用及机制。方法选取30只雄性C57小鼠,10只为正常对照组,其余20只心肌梗死造模后1周,予腹腔注射脂多糖30 min后,随机分为空白对照组及麝香保心丸干预组,各10只。观察各组间小鼠提睾肌微循环差异(流速及白细胞黏附);流式分析中性粒细胞黏附分子CD11b的表达。结果与空白对照组比较,麝香保心丸干预组可显著改善微循环障碍,并能降低白细胞黏附至微循环内皮表面,差异有统计学意义(P<0.05);进一步分析发现,药物干预组小鼠外周血中性粒细胞表面黏附分子CD11b表达显著降低,差异有统计学意义(P<0.05)。结论麝香保心丸可显著改善小鼠心肌梗死后微循环障碍,这可能与其抑制中性粒细胞表面黏附分子CD11b表达而减少白细胞的黏附密切相关。Objective To explore the protective effects of Shexiang Baoxin pills(SXBXP)on microcirculation dysfunction in mice with myocardial infarction and its mechanism.Methods Thirty male C57 mice were used,of which 10 mice were chosen as normal control group and 20 mice were given myocardial infarct operation.After one week of operation,20 mice were intraperitoneal injected with LPS and then randomized into control group(n =10)and SXBXP group(n =10).The changes of microcirculation were recorded and investigated,such as microcirculation flow velocity and white blood cells' adhesion.The expression of CD11 b in neutrophil was analyzed with flow cytometry.Results Compared with control group,the microcirculation dysfunction was alleviated and the number of white blood cells' adhesion to endothelial cells was decreased in SXBXP group(P〈0.05).The expression of CD11 b in neutrophil was decreased in SXBXP group(P〈0.05).Conclusion SXBXP can improve the microcirculation dysfunction in mice with myocardial infarction,which might be partly attributed to inhibiting the expression of CD11 b in neutrophil.
