摘  要:采用加热回流、超声辅助、微波辅助以及加压热水等方法提取白头翁总皂苷,比较不同提取方法对白头翁皂苷提取率、抑菌活性、抗氧化活性的影响;同时采用电镜扫描观察提取方法对药材结构的影响;在此基础上,研究加压提取的传质动力学。结果表明:采用加压热水方法提取白头翁皂苷具有提取率高(提取率6.74%,质量分数)、提取物抗氧化、抑菌活性强的特点;电镜扫描表明采用加压热水进行提取时,不破坏药材的细胞结构;加压热水提取的传质过程符合二阶动力学模型,其动力学方程为ρ_t=t[(2.6exp(-331.04/T))^(-1)+t(0.007 8T^2-6.416 9T+1 329.3)^(-1)]^(-1),提取过程活化能Ea为74.376 2 k J/mol。Triterpenoid glycosides from Radix Pulsatillae were extracted by hot water extraction,ultrasonic assisted extraction,microwave assisted extraction,and pressurized hot water extraction(PHWE).The extraction yield of triterpenoid glycosides as well as antibacterial and antioxidant activity of the extracts from four methods were compared.Scanning electron microscope images of the plant cells after four treatments were obtained to provide visual evidence of the extraction effect.The results show that the extraction yield of triterpenoid glycosides by PHWE was the highest,and the extracts obtained with PHWE also showed the higher antibacterial and antioxidant activity.The extraction model of a second-order mechanism was developed to predict the concentration of triterpenoid glycosides under various extraction temperatures.The extraction kinetic equation was described as ρ_t=t[(2.6exp(-331.04/T))^(-1)+t(0.0078T^2-6.4169T+1329.3)^(-1)]^(-1),the E_a was7.43762kJ/mol.
