摘  要:目的:探讨益气养阴祛瘀方对干燥综合征样非肥胖糖尿病(NOD)小鼠PBMC中TLR-IFNα通道表达的影响。方法:将30只NOD自发性干燥综合征(SS)样小鼠随机分为模型组、羟氯喹组、益气养阴祛瘀方组(中药组),每组10只。ICR小鼠10只为正常组。正常组和模型组给予生理盐水,羟氯喹组给予羟氯喹溶液75 mg/kg,中药组予益气养阴祛瘀方药液27.4 g/kg,观察小鼠的饮水量指标的变化,实时定量PCR技术检测TLR5和IFN-α mRNA表达水平。结果:与模型组相比,羟氯喹组和益气养阴祛瘀方组的饮水量有明显改善(P<0.05);模型组IFN-α mRNA相对表达量较正常组显著升高(P<0.05);与模型组比较,羟氯喹组、中药组IFN-α mRNA相对表达量明显降低(P<0.05);中药组IFN-α mRNA相对表达量低于羟氯喹组(P<0.05),模型组TLR5 mRNA相对表达量较正常组显著升高(P<0.05),羟氯喹组和中药组TLR5 mRNA相对表达量均低于模型组(P<0.05),但比正常组高(P<0.05)。结论:益气养阴祛瘀方对干燥综合征的治疗有效,推测与其调节NOD小鼠TLR-IFNα通路的表达有关。Objective:To investigate the effect of strengthening qi,nourishing yin and removing blood stasis formula on regulating TLR-IFNα signal pathway in NOD mice. Methods:The 30 NOD spontaneously sjogren′s syndrome(SS)samples of mice were randomly divided into model group,hydroxychloroquine group and Yiqiyangyin Stasis group. 10 rats were taken as normal control group. The mice in Chinese Herbs group were given 27.4 g/kg of Yiqiyangyin Quyu formula Decoction every day. The mice in Hydroxychloroquine group were given hydroxychloroquine 75 mg/kg by gavage. Mice of normal group and model group were given nomal saline. To observe the mice drinking. Changes in body mass index.Expression of IFN-α mRNA and TLR5 mRNA in peripheral blood mononuclear cells were detected using reverse transcriptase polymerase chain reaction(RT-PCR). Results:Compared with model group,water intake and hydroxychloroquine group Yiqiyangyin Stasis group improved significantly(P 0.05),model group,IFN-α mRNA relative expression was significantly higher than the normal group(P 0.05),compared with model group,hydroxychloroquine group,TCM group IFN-α mRNA relative expression levels were significantly lower(P〈0.05),compared with the normal group,there was a significant difference(P〈0.05),the Medicines IFN-α mRNA relative The expression lower than hydroxychloroquine group(P〈0.05). Model group,the relative expression of TLR5 mRNA were significantly higher than the normal group(P〈0.05),hydroxychloroquine group and Chinese medicine group TLR5 mRNA expression levels relatively lower than the model group(P 0.05),but higher than the normal group(P〈0.05). Conclusion:Strengthening Qi,Nourishing Yin and Removing Blood Stasis formula can be effective for the treatment of sjogren′s syndrome,presumably with lower expression in NOD mice TLR-IFNα pathway are related.
