摘 要:目的比较局部麻醉药丁卡因、布比卡因、罗哌卡因、普鲁卡因和利多卡因对神经细胞的毒性作用。方法分别采用不同浓度的上述5个局部麻醉药刺激体外培养的人神经细胞母细胞瘤SH-SY5Y细胞,构建局部麻醉药神经毒性的体外培养模型;未行任何刺激的细胞作为对照组。药物作用24h后,采用MTT法测定细胞存活率,显微镜下观察细胞形态,计算5个局部麻醉药的半数致死量。结果与对照组细胞比较,5个局部麻醉药均呈浓度依赖性降低SH-SY5Y细胞存活率(P<0.01)。半数致死量:丁卡因为212.7μmol/L,布比卡因为989.1μmol/L,罗哌卡因为3.6mmol/L,普鲁卡因为3.7mmol/L,利多卡因为7.4 mmol/L。结论局部麻醉药呈浓度依赖性诱导神经细胞毒性;其毒性大小依次为丁卡因>布比卡因>罗哌卡因>普鲁卡因>利多卡因。Objective To compare the neurotoxicity of local anesthetics tetracaine,bupivacaine,ropivacaine,procaine and lidocaine on nerve cells.Methods The human neuroblastoma SH-SY5 Ycells cultured in vitro were treated in vitro with different concentrations of tetracaine,bupivacaine,ropivacaine,procaine and lidocaine for 24 hours.The cells without drug stimulation were taken as the controls.The neurotoxicity was evaluated by MTT assay and morphological changes of the cells were observed under microscope.The median lethal doses(LD50)for 5 drugs were calculated.Results Compared with control cells,the viabilities of SH-SY5 Ycells treated with different concentrations of5 local anesthetics were all depreased dose-dependently(P〈0.01).The LD50 was 212.7μmol/L for tetracaine,989.1μmol/L for bupivacaine,3.6mmol/L for ropivacaine,3.7mmol/L for procaine,and7.4mmol/L for lidocaine.Conclusion The local anesthetics can induce the neurotoxicity in a dosedependent manner,which is in an order of tetracainebupivacaineropivacaineprocainelidocaine.
