不同中医治法对功能性勃起功能障碍模型大鼠eNOS mRNA、Cx43 mRNA表达的影响

摘　　要：目的:基于中医"肝藏血、主疏泄"藏象理论,研究不同治法对功能性勃起功能障碍模型大鼠阴茎海绵体eNOS mRNA、Cx43 mRNA表达的影响。方法:选取经交配后证实有正常勃起功能的雄性SD大鼠50只,随机分为空白组、模型组、补血活血法组、疏肝理气法组、疏肝理气活血法组,共五组,每组10只,采用尾部放血和悬吊激惹方法配合低铁饮食制备肝郁血虚勃起功能障碍模型,造模的第1d开始,在悬吊结束后1h,养血法组、疏肝法组与疏肝养血法组分别给予灌胃四物汤煎剂3.75g/kg、逍遥散煎剂9.22g/kg、四物汤与逍遥散合方煎剂12.96g/kg,空白组、模型组给予灌胃等体积生理盐水,1次/d,连续14d,末次灌胃24h后处死大鼠,取阴茎海绵体组织,HE染色观察阴茎海绵体组织病理学,PCR观察阴茎海绵体eNOS mRNA、Cx43 mRNA。结果:各组阴茎海绵体组织病理学改变均不明显;与空白组相比,模型组阴茎平滑肌组织Cx43mRNA、eNOS mRNA表达明显较低(P<0.05);与模型组相比,疏肝理气活血法组能显著提高阴茎平滑肌组织中Cx43 mRNA、eNOS mRNA表达(P<0.05),疏肝理气法组可显著提高阴茎平滑肌组织中Cx43 mRNA的表达(P<0.05),补血活血法组阴茎平滑肌组织中Cx43 mRNA、eNOS mRNA表达无统计学意义(P>0.05)。结论:疏肝理气活血和疏肝理气治法方药能够提高Cx43、eNOS活性,而补血活血治法方药不能提高Cx43、eNOS活性。Objectives： To investigate the effects of different therapeutic methods on expression of e NOS mRNA and Cx43 mRNA in model rats with functional erectile dysfunction based on theory of liver storing blood and governing regulating. Methods： 50 SD male rats with normal sexual function were selected by mating experiment and were randomly divided into blank group,model group,nourishing blood group,smoothing liver-regulating Qi group and nourishing blood and smoothing liver-regulating Qi group,with 10 rats in each group. With the tail bloodletting and suspensory irritability combined with low iron content diet,erectile dysfunction rats with stagnation of the liver-qi and blood-deficiency were bred. In the first day of molding,after 1 hour of suspensory,nourishing blood group,smoothing liver-regulating Qi group and nourishing blood and smoothing liver-regulating Qi group were given3. 75g/kg gavage Siwu soup,9. 22g/kg Xiaoyaosan decoctum and 12. 96g/kg combined decoctum of Siwu soup and Xiaoyaosan respectively. Blank group and model group were given gavage isopyknic physiological saline,once a day for14 days. All rates were killed 24 hours after the last gavage. Corpus cavernosum of the rates was observed pathologi-cally with HE staining. The e NOS mRNA and Cx43 mRNA were observed with PCR. Results： The pathological changes of corpus cavernosum were unobvious. The expression of e NOS mRNA and Cx43 mRNA in penis smooth muscle of model group was declined significantly comparing with blank group. Comparing with model group,the expression of e NOS mRNA and Cx43 mRNA in nourishing blood and smoothing liver-regulating Qi group increased with statistical significance（ P〈0. 05）. The expression of Cx43 mRNA was significantly increased in smoothing liver-regulating Qi group（ P〈0. 05）. But the expression of Cx43 mRNA and e NOS mRNA in nourishing blood group had no significant change（ P〉0. 05）.
