摘  要:目的探讨Notch信号通路对脂肪间充质干细胞向心肌细胞分化的影响及分子机制。方法将分离培养的脂肪间充质干细胞利用5-氮杂胞苷(5-azacytidine,5-aza)体外进行定向诱导,Jagged1组加入Notch激活剂Jagged1,DAPT组加入Notch激活剂Jagged1和抑制剂DAPT,对照组加入PBS缓冲液。用荧光定量PCR、免疫组化、Western Blot等方法检测干细胞分化为心肌细胞的情况及Notch信号系统的表达。结果在体外,5-aza诱导后,脂肪间充质干细胞可以向心肌细胞分化,Jagged1组的干细胞与DAPT组及对照组相比,其细胞分化比率明显提高,并且其心肌标志物Tn T的表达量也增多,Notch1和DTX1表达量增强。结论 Notch信号系统对脂肪间充质干细胞分化为心肌细胞起正向调控作用。Objective To investigate the regulation of differentiation of Adipose derived mesenchymal stem cells (ADMSCs) into cardiomyocytes by Notch signaling system. Method ADMSCs were isolated and cultured from adult adipose tissue and induced with 5-azacytidine to differentiate to cardiomyocytes. Jagged1, an activator of the Notch signaling system, was added into the culture medium of the experiment group. The control group was added with an inhibitor of the Notch signaling system. The cells were cultured in the medium added with phosphate-buffered saline used as blank control group. Then the cultured cells were collected. The expression of Notch signal and TnT was detected by RT-PCR, immunohistochemistry and Western Blot. Result ADMSCs can differentiate into cardiomyocytes in vitro. Compared with the two control groups, the differentiate percentage was higher in the experimental group. RT-PCR and Western Blot showed that the expression level of the receptor Notch l and DTX1 was significantly higher. Immunohistochemistry and Western Blot showed a significantly increased expression of TnT. Conclusion When the Notch signaling system is activated, the differentiation of ADMSCs into cardiomyocytes enhances, while the effect can be inhibited by DAPT.
