摘  要:目的:探讨核苷类逆转录酶抑制剂(NRTIs)对大鼠卵母细胞成熟度及受精能力的影响。方法:采用NRTIs药物(AZT和3TC)灌饲雌性大鼠12个月,在用药3、6、9及12个月分别处死大鼠,以酶联免疫吸附试验法检测大鼠卵母细胞成熟相关因子的表达变化。同时观察大鼠卵母细胞体外培养成熟度及胚胎发育潜能的变化。结果:NRTIs给药后大鼠卵母细胞的成熟度受到显著影响,卵裂率和囊胚形成率均下降(P<0.05)。卵母细胞成熟相关因子MPF等的表达水平也显著下降(P<0.05)。结论:NRTIs影响大鼠卵母细胞的成熟及胚胎发育潜能,降低其生育能力。Objective: To investigate the effect of Nucleoside Reverse Transcriptase Inhibitors (NRTIs, such as AZT and 3TC) on the mice oocyte maturity and fertile potentiality. Methods and materials.. After the female mice fed with AZT and 3TC, they were killed at 3, 6, 9 and 12 months, and the factors related to ovum maturation were detected by the method of enzymeqinked immunosorbent assay (ELISA). The changes of mice ovum maturation and the embryo developmental potentiality in-vitro culture were observed at the same time. Results: The mice ovum maturity was negatively influenced by AZT and 3TC, and the rate of embryo cleavage and blastocyst were decreased significantly after AZT and 3TC fed (P〈0.05). Conclusions: The ovum maturity and the embryo development potentiality were negatively influenced by AZT and 3TC, and the fecundity of mice decreased significantly after AZT and 3TC fed.
