摘 要:目的:观察茜草炭不同提取物对子宫出血模型大鼠的影响,筛选茜草炭止血作用的最佳提取方式。方法:空白组与其余各组分别灌胃给予0.5%CMC-Na、戊酸雌二醇(2.5 mg·kg^(-1)),连续灌胃3周,以宫血宁为阳性药,对照组及模型组静脉滴注羧甲基纤维素钠(CMC-Na),其他各组静脉滴注给予相应的治疗药物,观察茜草炭醇提物、复合提取物、醇提后水提物以及水煎物对子宫出血模型大鼠血液流变学、血小板系统以及纤溶系统的影响。结果:与空白组比较,模型组大鼠低切变率下的全血黏度降低(P<0.05),TT、PT、APTT显著延长(P<0.05、0.01),FIB、6-keto-PGF1α含量显著增加(P<0.01),TXB2和6-keto-PGF1α/TXB2显著降低(P<0.05、0.01)。与模型组比较,茜草炭复合提取物可显著升高各切变率下全血黏度(P<0.05、0.01),水煎物显著提高低切变率下全血黏度(P<0.05);醇提物、复合提取物和醇提后水提物显著缩短TT、PT、APTT(P<0.05、0.01),水煎物显著降低APTT(P<0.05),醇提物和复合提取物显著增加FIB含量(P<0.05),复合提取物和醇提后水提物显著降低6-keto-PGF1α含量(P<0.05、0.01),显著升高TXB2含量(P<0.05),复合提取物显著升高6-keto-PGF1α/TXB2比值(P<0.05),复合提取物有效缓解子宫内膜增厚、出血现象。结论:茜草炭复合提取物止血作用强于茜草炭其他提取物,即复合提取法为茜草炭止血作用的最佳提取方式。Objective: To observe the effect of different extracts from charred radix rubiae in dyfunctional uterine bleeding( DUB) model rats in order to screen the optimal extraction method of hemostasis effect. Methods: The SD rats were divided into seven groups. The rats in the control group were fed with CMC-Na for 3 weeks and the other groups were fed with estradiol valerate for 3 weeks. Gongxuening was taken as the positive control drug. The rats in the control group and model group were fed with CMC-Na for 7 days and the rats in other groups were fed with corresponding drugs simultaneously. The influence of the different polarities of carbonized R. cordifolia compound to dyfunctional uterine bleeding( DUB) rats' hemorheology,coagulation system and platelet system. Results: Compared with the blank group,the model group could reduce the low whole blood viscosity( P〈0. 05)and prolong the TT,PT and APTT( P〈0. 05,P〈0. 01),increase the FIB and 6-keto-PGF1αcontents,decrease the TXB2and6-keto-PGF1α/ TXB_2( P〈0. 05,P〈0. 01). Compared with the model group,compound extraction could significantly increase the whole blood viscosity( P〈0. 05,P〈0. 01). The water extract could increase the low blood viscosity( P〈0. 05). The alcohol extraction,the compound extraction and water extraction by alcohol extraction could remarkably reduce TT,PT and APTT( P〈0. 05,P〈0. 01). The water extraction reduced APTT obviously( P〈0. 05). The alcohol extraction and the compound extraction increased the FIB significantly( P〈0. 05). The compound extraction and water extraction by alcohol extraction could decrease 6-keto-PGF1αand increase TXB2( P〈0. 05,0. 01). The compound extraction could rise 6-keto-PGF1α/ TXB2( P〈0. 05).Besides,it could also relieve endometrial thickening and bleeding. Conclusion: The hemostatic effect of compound extraction is stronger than the others.In other words,the optimal extraction method of its hemostatic effect is the compound extraction
