摘 要:目的:研究玉竹甾体皂苷类成分及其对小鼠脾细胞增殖的影响。方法:利用各种色谱技术进行分离纯化,根据化合物的理化性质和光谱数据进行结构鉴定,应用MTT法考察玉竹正丁醇层总皂苷和3个皂苷类成分对小鼠脾细胞体外增殖的影响。结果:从玉竹中分离并鉴定了5个化合物,分别为β-谷甾醇(Ⅰ)、胡萝卜苷(Ⅱ)、无刺枣苷Ⅰ(Ⅲ)、26-O-β-D-吡喃葡萄糖基-22-O-甲基-(25S)-呋甾-5-烯-3β,26-二醇-3-O-β-D-吡喃葡萄糖基(1→2)-[β-D-吡喃木糖基-(1→3)]-β-D-吡喃葡萄糖基(1→4)β-D-吡喃半乳糖苷(Ⅳ)、26-O-β-D-吡喃葡萄糖基-(25S,25R)-呋甾-5-烯-3β,14α,22α,26-四醇-3-O-β-D-吡喃葡萄糖基(1→2)-[β-D-吡喃木糖基-(1→3)]-β-D-吡喃葡萄糖基(1→4)β-D-吡喃半乳糖(Ⅴ)。小鼠脾细胞体外增殖活性研究发现玉竹正丁醇层总皂苷、化合物Ⅲ、Ⅳ、Ⅴ具有明显的促进小鼠脾细胞增殖的活性。结论:化合物Ⅳ、Ⅴ为首次从该种内分离得到,化合物Ⅲ为首次从该属植物中分离得到,玉竹中皂苷类成分是玉竹免疫调节作用的物质基础之一。Objective: To study the saponins of Polygonatum. odoratum( Mill.) Druce and its effect on murine splenocytes proliferation. Methods: The chemical constituents were isolated by column chromatography on silica gel,RP silica gel,and so on. The structures were identified by spectral data and chemical methods. The method of MTT was used to examine to the effects of three saponins and total saponins of n-Bu OH fraction. Results: 5 compounds were isolated and identified as β-sitosterol( Ⅰ),Daucosterol( Ⅱ),Zizybeoside IⅠ( Ⅲ),26-O-β-D-glucopyranosyl-22-O-methyl-( 25S)-furost-5-ene-3β,26-diol-3-O-β-D-glucopyranosyl( 1→2)-[β-D-xylopyranosyl-( 1→3) ]-β-D-glucopyranosyl( 1→4) β-D-galactopyranoside( Ⅳ),26-O-β-D-glucopyranosyl-( 25 S,25R)-furost-5-ene-3β,14α,22α,26-quarol-3-O-β-D-glucopyranosyl( 1→2)-[β-D-xylopyranosyl-( 1→3) ]-β-D-glucopyranosyl( 1 →4) β-D-galactopyranoside( Ⅴ). By the experiment of murine splenocytes proliferation acitivty in vitro,the results showed that the total saponins of n-Bu OH fraction,3 saponins had promoted the proliferation of mouse spleen cells. Conclusion: Compound Ⅳ、Ⅴ is obtained from this plant for the first time,compound Ⅲ is obtained from Polygonatum for the first time. The total saponins is one of the material basis of immunomodulatory effects of Polygonatum. odoratum( Mill.) Druce.
