摘  要:试验旨在探讨蛋白激酶CLK1在早期鸡胚胎发育过程中的表达情况。应用分子克隆技术构建CLK1/p MD18-T重组质粒,制备地高辛标记的CLK1 RNA原位杂交探针,利用Real-time PCR和原位杂交技术检测CLK1 mRNA在早期鸡胚胎发育中的表达及分布。Real-time PCR结果显示,CLK1 mRNA在早期鸡胚各时期中均有不同程度的表达;原位杂交结果显示,CLK1的mRNA在鸡胚的不同时期不同部位均有不同程度的表达,且在头部和尾部表达明显。本研究阐明了CLK1在早期鸡胚胎发育过程中的表达及分布,为进一步研究CLK1在鸡胚中的功能及作用机制奠定基础。To analyze the expression pattern of CDK-like kinase 1(CLK1) in the early chick embryos,the plasmid of CLK1/p MD18-T was constructed and the digoxigenin-labeled sense and antisense RNA probes were produced by in vitro transcription according to a standard protocol.The expression pattern of CLK1 in the early chick embryos was investigated using Real-time PCR and in situ hybridization with the synthesized probes.The results of Real-time RCR showed that mRNA of CLK1 had different degrees of expression in the different periods of the early chick embryos.The results of in situ hybridization showed that mRNA of CLK1 was expressed inordinately in different parts of the early chick embryos,especially in head and tail.The expression pattern of CLK1 in early chick embryos obtained in this study provides an approach to further investigate the biological functions of CLK1 under physiological and pathological conditions.
