摘  要:为了解伪狂犬病病毒(PRV)和猪圆环病毒2型(PCV2)感染仔猪后不同时间点的病毒载量及排毒规律,将34头健康仔猪分为4组,第1~3组分别为人工接种PRV、PCV2和PRV+PCV2组,第4组为空白对照组。分别在接种后第3、7、14、21、35天采集试验仔猪的鼻拭子和肛门拭子,用Taq Man荧光定量PCR(q PCR)对其进行定量检测,并分析仔猪排毒的变化规律。结果,接种后第3天,3个试验组仔猪肛门拭子、鼻拭子中均分离到PRV或PCV2;其中PRV单独感染后的病毒载量在第14天达到峰值331.67 copies/L;PCV2单独感染后的病毒载量在第3天达到峰值10 742.01 copies/L;PRV+PCV2混合感染后鼻拭子和肛门拭子的PRV分别在第14天和第7天达峰值980.96 copies/L和2 230.056 copies/L;鼻拭子和肛门拭子PCV2在感染后第14天达到峰值69.413 copies/L和271.72 copies/L;混合感染仔猪于感染后第8~14天发生死亡。3个感染组之间无论是肛门拭子还是鼻拭子,PRV、PCV2病毒载量均差异显著(P<0.05)。上述结果表明,PRV单独感染、PCV2单独感染和二者混合感染均对仔猪造成严重危害,除引起仔猪发病死亡外,这2种病毒均能通过呼吸道和消化道向外排毒。In order to determine the dynamics of pseudorabies virus(PRV)and porcine circovirus type2(PCV2)loading and shedding,34 healthy piglets were divided into 4 groups in this study.The groups 1,2and 3 were inoculated with PRV,PCV2 and PRV+PCV2,respectively,while group 4 was set as control group.The nasal swab was collected and tested for viruses on the 3rd,7th,14 th,21th and 35 th days after inoculation.The q PCR method was used for quantitative detection of the changes of viruses.In result,PRV and PCV2 were detected from anal swab and nasal swab of the piglets in the 3 experimental groups on3 rd day post-inoculation.In the PRV infection group,the viral load reached the peak of 331.67 copies/L on 14 th day and in the PCV2 infection group,reached a peak value of 10 742.01 copies/L on the 3rd day,after inoculation.PRV reached the peak of 980.96 copies/L and 2 230.056 copies/μL on the 14 th and 7th day and PCV2 reached the peak of 69.413 and 271.72 copies/L on the 14 th day after inoculation.The piglets died from 8th to 14 th day post-inoculation in the PCV2+PRV co-infection group. The level of PRV and PCV2 in the anal swab and the nasal swab were significantly different among the 3 experimental groups(P〈0.05). In conclusion, both the PRV or PCV2 single infection and the co-infection seriously harm the piglets,lead the death of piglets and result in shedding the 2 viruses through respiratory tract and digestive tract.
