摘  要:目的探讨矮膜苞芹水提物对两肾一夹型肾血管性高血压大鼠(Renal hypertensive rats,RHR)的降血压作用及其机制。方法利用肾动脉狭窄法建立两肾一夹高血压大鼠模型,将大鼠随机分为假手术组、模型组、阳性对照药卡托普利组及矮膜苞芹水提物高、低剂量组,灌胃给药6w,每周无创微动脉测压法测量大鼠尾动脉血压,6w后测定血浆中血管紧张素转化酶(ACE)、醛固酮(ALD)、抗氧化酶超氧化物岐化酶(SOD)、一氧化氮(NO)、内皮素-1(ET-1)、血管紧张素Ⅱ(AngⅡ)含量,并观察其肾脏、心脏组织形态学变化。结果与模型组比较,矮膜苞芹水提物能降低RHR大鼠血压,差异有统计学意义(P<0.01);与卡托普利相比,矮膜苞芹水提物高剂量组降血压作用与卡托普利组相当,差异无统计学意义(P>0.05);与模型组相比,矮膜苞芹水提物能降低RHR大鼠血浆中ACE(P<0.01)、ALD(P<0.01)、ET-1(P<0.01)、AngⅡ(P<0.05),能升高RHR大鼠血浆中SOD活力(P<0.01)。矮膜苞芹水提物在某种程度上对RHR靶器官肾脏和心脏的损害有逆转作用。结论矮膜苞芹在防治肾性高血压方面具有独特的优势及潜在的应用前景。Objective To investigate the effects of water extract from Hymenoleana nanaon Renal hypertensive rats.Methods Two-kidney-one-clip RHRs were obtained by narrowing left kidney arteries.5groups were randomly divided into sham-operation group,model group,positive control captopril group,water extract fromHymenoleana nanalow-dose and high-dose groups.The drugs were given by intragastric administration for 6weeks.Systolic blood pressure(SBP)was measured by tail cuff approach every week.After the sixth week,the ACE,ALD,SOD,NO,ET-1and AngⅡ contents in the plasma were measured.Then,RHR kidney and heart HE staining pathological changes were observed.Results Compared with control group,water extract from Hymenoleana nanacan significantly lower SBP in RHR(P〈0.01);Compared with captopril group,high dose water extract from Hymenoleana nana group was as effective as captopril group but without significance(P〉0.05).water extract from Hymenoleana nana can alleviate ACE(P〈0.01),ALD(P〈0.01),ET-1(P〈0.01),AngⅡcontents(P〈0.05),and improve activity determination of SOD(P〈0.01)in the plasma.To some extent,water extract from Hymenoleana nana had the role to reverse the RHR target organ(kidney and heart)damage.Conclusion Hymenoleana nanais a potential promising therapeutic agent for renal hypertensive.
