摘 要:目的:了解腹腔注射SMAD3抑制剂柚皮素对小鼠骨骼肌急性钝挫伤后纤维化的影响。方法:72只7~8周龄C57bl/6小鼠(20~24 g)随机分为4组,每组18只。包括正常对照组(A组)、普通损伤组(B组)、损伤后二甲基亚砜(DMSO)注射组(C组)、损伤后柚皮素注射组(D组)。B、C、D三组均用自制打击装置制造右侧胫骨前肌中段急性钝挫伤模型;C、D组分别腹腔注射单纯DMSO溶液、柚皮素DMSO溶液,1次/天,直至伤后28天取材。所有小鼠处死取材后,Western Blot方法检测4组小鼠右胫骨前肌SMAD3、p SMAD3、CollagenⅠ、α-SMA的表达;HE及Masson染色评估胫骨前肌的组织学变化;电生理学方法检测各组小鼠胫骨前肌快速颤搐收缩和强直收缩能力,以了解小鼠损伤骨骼肌力学性能恢复情况。结果:1)B、C、D组所有损伤后小鼠骨骼肌中SMAD3表达均高于A组,B、C组p SMAD3表达升高,D组SMAD3以及p SMAD3的表达低于B、C组;2)B、C、D组中CollagenⅠ与α-SMA水平高于A组,但D组的CollagenⅠ与α-SMA水平低于B、C组;3)HE染色发现,损伤骨骼肌中间质较正常组增多,Masson染色证实B、C、D组急性钝挫伤后骨骼肌出现明显纤维化,但D组的骨骼肌纤维化面积显著低于B组和C组;4)损伤后骨骼肌快速颤搐收缩与强直收缩能力显著下降,但D组明显优于B、C组。结论:SMAD3抑制剂柚皮素可抑制骨骼肌损伤后的SMAD3磷酸化水平,改善纤维化,减轻疤痕生成,从而促进骨骼肌损伤修复,提高骨骼肌损伤后的愈合质量。Objectives To understand the effect of intraperitoneal injection of naringenin,a SMAD3 in-hibitor,on the skeletal muscle after acute contusion in a mouse model. Methods Seventy-two mice of 7-8 weeks old(20-24 g)were randomly divided into a control group,an acute contusion(B)group,anacute contusion+1%DMSO injection(C)group and an acute contusion+naringenin injection(D)group,each of 18. The acute contusion model was created by hitting the right tibialis anterior muscle inmice of all groups except the control group. Intraperitoneal injection of 1%DMSO and naringenin weregiven to group C and D respectively every day until execution,while the 18 mice in the control groupwere fed without injury or injection. The time of injury was set as Day 0. After being fed for 28 days,all mice were executed and the right tibialis anterior was harvested. Western blotting was used to de-tect the difference of SMAD3,p SMAD3,Collagen Ⅰ,and α-SMA expression among the 3 groups. Hematoxylin-Eosin(HE)staining and Masson staining were used to detect the difference of pathologicalchanges. Moreover,the appearance of fast twitch contraction and tetanic contraction were also document-ed to figure out the quality of the injured skeletal muscle. Results Compared with the control group,the SMAD3 and p SMAD3 level in injured skeletal muscle increased,but both were less in group Dthan group B and C. Similarly,the average level of Collagen Ⅰ and α-SMA in all three injury groupswas higher than the control group,but the level of these indexes were lower in group D than that ingroup B and C. HE staining showed more mesenchyme in injury groups than the control group. Mas-son staining found the upregulation of fibrosis in injured muscles,with the area of fibrosis in group Dsignificantly lower than group B and C. Compared with control,the injured skeletal muscle had signifi-cantly poorer fast twitch and tetanic contraction performance,with the condition of group D significantlybetter than group B and C.
