摘 要:目的观察橙皮苷对IgA肾病大鼠模型肾脏组织JAK/STAT信号通路蛋白STAT3和p-STAT3表达的影响。方法将60只SD大鼠随机分为对照组、模型组以及治疗组各20只,模型建立成功后,治疗组给予橙皮苷灌胃治疗,对照组和模型组给予相应生理盐水灌胃,分别于治疗4周和8周后处死大鼠,收集血清、尿液和肾脏组织,检测其肾功能、氧化应激相关指标以及肾脏组织STAT3和p-STAT3蛋白及mRNA的表达特点。结果治疗4、8周后模型组和治疗组肌酐(Cr)、尿素氮(BUN)、24 h尿蛋白及STAT3、p-STAT3蛋白和mRNA表达明显高于对照组(P<0.05);且治疗组BUN、24 h尿蛋白和STAT3 mRNA表达明显低于模型组(P<0.05)。治疗8周后治疗组Cr水平、STAT3和p-STAT3蛋白表达明显低于模型组(P<0.05)。治疗4周后模型组和治疗组超氧化物歧化酶(SOD)低于对照组,丙二醛(MDA)高于对照组(P<0.05)。治疗8周后,模型组SOD低于对照组,MDA高于对照组(P<0.05)。治疗4周和8周后治疗组SOD高于模型组,MDA低于模型组(P<0.05)。结论橙皮苷对IgA肾病大鼠肾功能具有改善作用,其机理可能与提高抗氧化能力,降低STAT3蛋白的表达有关。Objective To observe effect of Hesperidin on signal transducer and activator of transcription 3 ( STAT3 ) and p-STAT3 protein expressions related to IAK (janus kinase)/STAT signal in kidney tissues of IgA nephrop- athy model rats. Methods A total of 60 SD rats were randomly divided into control, model and treatment groups (n= 20 for each group). After models were successfully established, treatment group was treated with Hesperidin, and model group and control group were lavaged with corresponding normal saline respectively, and then rats were sacrificed in 4th and 8th weeks of treatment, and serum, urine and kidney tissues were collected. Renal function, related indexes of oxida- tive stress and protein and mRNA expression characteristics of STAT3 and p-STAT3 in kidney tissues were detected. Re- suits In 4th and 8th weeks of treatment, levels of creatinine (Cr) , blood urea nitrogen (BUN) , 24 h urinary protein and protein and mRNA expressions of STAT3, p-STAT3 in model and the treatment groups were significantly higher than those in control group (P 〈 0. 05) ; levels of BUN, 24 h urinary protein and STAT3 mRNA expression in treatment group were significantly lower than those in model group (P 〈 0. 05). In 8th weeks of treatment, level of Cr, STAT3 and p-STAT3 protein expressions in treatment group were significantly lower than those in model group ( P 〈 0.05 ). In 4'h weeks of treatment, superoxide dismutase (SOD) levels were significantly lower, malondialdehyde (MDA) levels were significant- ly higher in model and the treatment groups than those in control group (P 〈 0.05). In 8th weeks of treatment, SOD was significantly lower, while MDA level was significantly higher in model group than those in control group (P 〈 0. 05 ). In 4th and 8th weeks of treatment, SOD level was significantly higher, while MDA level was significantly lower in treatment group than those in model group (P 〈 0.05). Conclusion Hesperidin can improve renal function of IgA
