龙须藤总黄酮对大鼠骨关节炎治疗作用及机制

摘　　要：目的考察龙须藤总黄酮对骨关节炎动物模型的治疗作用和机制。方法釆用4%木瓜蛋白酶关节腔内注射法建立骨关节炎大鼠模型。以大鼠关节直径变化,足趾容积变化、关节病理切片及骨关节Makin's评分为指标,观察龙须藤总黄酮对膝关节炎大鼠的影响。利用ELISA法检测大鼠血清中IL-1β、TNF-α含量,初步探讨龙须藤总黄酮治疗骨关节炎的作用机制。结果与模型组相比,龙须藤总黄酮各组均能不同程度地使骨关节大鼠关节直径、足趾容积、Makin's评分降低,并能显著降低大鼠血清中IL-1β和TNF-α的含量。结论龙须藤总黄酮对膝关节炎大鼠具有治疗作用,其抗炎机制可能与其抑制IL-1β和TNF-α分泌有关。Objective To study the therapeutic effect and mechanism of total flavones from Bauhinia championii on rats with osteoarthritis（ OA） . Methods The OA rat model was established by intra-articular injection of papain. The changes of knee diameter, paw volume and histopathology were measured. The level of IL-β and TNF-α was detected by ELISA. Results Compared with the model group, the changes of knee diameter, paw volume and Makin′s score were decreased in OA rats treated with total flavones from Bauhinia championii. Meantime, the contents of serum IL-1β and TNF-α were also reduced in total flavones-treated OA rats. Conclusion The total flavones derived from Bauhinia championii exhibits a therapeutic effect on OA rats, which may be related to inhibition of IL-1β and TNF-α production.
