摘  要:热激蛋白90α(Heat shock protein 90,Hsp90α)在传染性法氏囊病病毒(Infectious bursal disease,IBDV)的致病过程中发挥重要作用。蛋白互作及其亲和力均可能导致不同的信号转导和生物效应,鸡源Hsp90α与IBDV衣壳蛋白VP2的互作已经被证明,但两者之间的亲和力尚未被鉴定。研究通过表达和纯化鸡的Hsp90α和IBDV超强毒株的VP2蛋白,运用表面等离子共振(Surface plasmon resonance,SPR)技术,揭示Hsp90α和IBDV VP2蛋白两者互作的动态过程,并确定其亲和力((KD)为7.105×10^(-8)M),为强相互作用。研究结果对进一步揭示IBDV的致病机制有重要意义。Heat shock protein 90 alpha(Hsp90α)plays important roles in the infection of infectious bursal disease virus(IBDV). Interactions between proteins and their affinity may induce different signal transductions and biological effects,and the interaction between chicken Hsp90α and capsid protein VP2 of IBDV has been identified,but their affinity is unknown. To identify the affinity between Hsp90α and VP2 of very virulent IBDV,these two proteins were expressed and purified,and then their affinity was identified with surface plasmon resonance(SPR). This study disclosed the dynamic process of Hsp90α-CK1α interaction,and the high affinity was identified(KD=7.105×10^-8M),which would be beneficial for further understanding the pathogenic mechanism of IBDV.
