摘 要:目的应用超高压液相-四极杆-飞行时间串联质谱(UPLC-Q-TOF-MS/MS)技术的代谢组学方法探索补气药干预气虚大鼠的生物学基础。方法采用限食+力竭游泳的方法建立气虚大鼠模型,运用Agilent 6538UPLC-Q-TOF仪对空白组、气虚模型组、人参高剂量组(0.324 g·kg^(-1))、人参低剂量组(0.162 g·kg^(-1))、黄芪高剂量组(1.08 g·kg^(-1))、黄芪低剂量组(0.54 g·kg^(-1))各组大鼠血浆代谢物进行检测。结果 MPP软件分析与Metlin数据库检索寻找到与机体代谢密切相关的吲哚乙酸、溶血磷脂酰胆碱、十八碳酰胺、十八碳烯酸、脱氧胞苷等代谢差异物。结论本研究能够有效地探索补气药干预气虚大鼠作用的相关生物学基础。Objective To explore the biological basis of intervention effect of Qi-tonifying herbs on Qi-deficiency rats with metabolomic method based on ultra-performance liquid chromatography-quadrupole-time-of-flight tandem mass spectrometry(UPLC-Q-TOF-MS/MS)technology. Methods The Qi-deficiency rat model was established with caloric restriction combined with excessive swimming. Plasma from the rats of blank group,model group, high-dose Radix Ginseng group(0.324 g·kg(-1)),low-dose Radix Ginseng group(0.162 g·kg(-1)),high-dose Radix Astragali group(1.08 g·kg(-1)),low-dose Radix Astragali group(0.54 g·kg(-1)) group was detected by Agilent 6538 UPLC-Q-TOF. Results Indoleacetic acid,lysophosphatidylcholine,octadecanamide,vaccenic acid,deoxycytidine and so on,which were closely related to the metabolism in the body,were obtained after Metline database retrieval and analysis with MPP software. Conclusion The results will supply evidence for the exploration of related biological basis of intervention effect of Qi-tonifying herbs on Qi-deficiency rats.
