摘 要:目的建立HPLC法测定清热暗疮丸中绿原酸、栀子苷、穿心莲内酯和脱水穿心莲内酯的含量。方法色谱柱为Agilent Eclipse XDB-C_(18)(4.6 mm×150 mm,5μm);流动相为乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱;流速为1.0 m L·min^(-1);柱温:30℃;波长切换时间序列采样:0~7 min为327 nm,7~12 min为238 nm,12~19 min为225 nm,19~40 min为254 nm。结果绿原酸、栀子苷、穿心莲内酯和脱水穿心莲内酯分别在0.188~3.754μg(r=0.999 8)、0.164~3.272μg(r=0.999 8)、0.020~0.392(r=0.999 9)μg和0.039~0.777μg(r=0.999 9)范围内线性关系良好。平均回收率分别为99.53%,RSD为1.7%(n=9);99.33%,RSD为1.5%(n=9);99.59%,RSD为1.8%(n=9)和99.46%,RSD为1.5%(n=9)。5批清热暗疮丸中绿原酸、栀子苷、穿心莲内酯和脱水穿心莲内酯含量分别为9.48,8.05,0.92,1.92 mg·g^(-1)。结论该方法准确、快捷、可靠,可更好地用于清热暗疮丸的质量控制。Objective To establish a HPLC method for simultaneous determination of chlorogenic acid,geniposide,andrographolide and dehydroandrographolide in Qingre Anchuang Pills. Methods Agilent Eclipse XDB-C18(4.6×150mm,5 μm)column was used. The mobile phase was acetonitrile(A)-water containing 0.1 % phosphoric acid(B)with gradient elution mode at the flow rate of 1.0 m L·min-1. The detection wavelength was 327 nm(0-7 min)for chlorogenic acid,238 nm(7-12 min) for geniposide,225 nm(12-19 min) for andrographolide,254 nm(19-40 min) for dehydroandrographolide. Results Chlorogenic acid had a good linearity in the range of 0.188-3.754 μg, and the average recovery was 99.53 % with RSD being 1.7 %(n=9). Geniposide had a good linearity in the range of 0.164-3.272 μg,and the average recovery was 99.33 % with RSD being 1.5 %(n=9). Andrographolide had a good linearity in the range of 0.020-0.392 μg, and the average recovery was 99.59 % with RSD being 1.8 %(n=9).Dehydroandrographolide had a good linearity in the range of 0.039-0.777 μg,and the average recovery was 99.46 %with RSD being 1.5 %(n=9). The contents of chlorogenic acid,geniposide,andrographolide and dehydroandrographolide in 5 batches of Qingre Anchuang Pills were 9.48,8.05,0.92,1.92 mg·g(-1)respectively. Conclusion The established method is simple,accurate and reproducible. It can be used for the quality control of Qingre Anchuang Pills.
