生殖营养方多糖提取条件优化研究

摘　　要：目的:优化生殖营养方多糖的提取条件。方法:设计L_9(3~4)正交实验,采用苯酚-硫酸法显色,以多糖含量为指标,确定碱水提取生殖营养方中多糖的适宜条件。结果:碱水提取多糖的适宜条件:碱水pH=10~11,料液比为1∶70,提取1次,提取时间0.5h,醇沉乙醇浓度75%,该条件下多糖提取率为39.94%。结论:该法多糖提取率高,重现性好,又经济简单,可为生殖营养方多糖提取研究提供参考。Objective:The optimization of extraction conditions of polysaccharide from Shengzhi Yingyangfang.Methods:The L_9(3~4)orthogonal design was used to determine the optimanl extraction method.The content of polysaccharide was taken as an evaluation index with the method of phenol-H_2SO_4 for Shengzhi Yingyangfang.Results:The extraction rate of polysaccharide was 39.27%,When the pH of alkaline was 10~11,solid-liquid ratio was 1∶70,the times that polysaccharide was extracted was one time,the time of extraction was half of hour,and the ethanol concentration of 75% ethanol precipitation.Conclusion:This method was simple and economical and had good reproducibility,as the same time,the rate of polysaccharide extraction was high,which can provide reference for polysaccharide extraction of Shengzhi Yingyangfang.
