摘 要:目的探讨马齿苋生物碱对乳腺癌裸鼠肿瘤的抑制作用。方法利用体外培养高、低转移的人乳腺癌细胞株MDA-MB-231和MCF-7细胞,并将两组癌细胞体外培养后移植于裸鼠体内,观察马齿苋生物碱对肿瘤细胞的抑制效率。比较各组肿瘤组织瘤重,计算肿瘤生长抑制率;通过病理切片HE染色观察各组肿瘤组织病理学变化,并通过高效液相色谱分析肿瘤组织对马齿苋生物碱的亲和力。利用ELISA检测方法分析肿瘤局部组织VEGF的表达水平。结果马齿苋MDA-MB-231组和马齿苋MCF-7组肿瘤体积明显缩小,MCF-7组和MDA-MB-231组肿瘤体积较马齿苋干预组明显增大,差异有统计学意义(P=0.000)。马齿苋生物碱干预乳腺癌裸鼠后,病理显示肿瘤细胞减少,并伴有肿瘤坏死和调亡。实验结果显示肿瘤组织对马齿苋生物碱有较高的亲和力。同时研究还显示马齿苋生物碱对肿瘤的VEGF有显著的抑制作用(P=0.000)。结论马齿苋生物碱能有效抑制乳腺癌细胞的生长,调控肿瘤细胞代谢,可能有望成为乳腺癌的新的治疗方法和手段。Objective To study the inhibitory effect of alkaloids from purslane on breast cancer in nude mice. Methods Human breast cancer cell lines MDA-MB-231 with high metastatic rate and MCF-7 cells with low metastatic rate were cultured in vitro. And the two cancer cell lines were transplanted in nude mice after being cultured in vitro. The inhibitory effect of purslane alkaloids on tumor cells was observed. The tumor growth inhibition rate was calculated by comparing tumor weight of tumor tissue. The histopathological changes of the tumor tissues were observed by HE staining, and the affinity of the tumor tissue to the alkaloid of the purslane was analyzed by high performance liquid chromatography (RP-HPLC). The expression level of VEGF in local tissue was analyzed by ELISA. Results The tumor vol- ume of purslane MDA-MB-231 group and purslane MCF-7 group was significantly reduced, and the tumor volume of MCF-7 group and MDA-MB-231 group was significantly higher than that of the control group, and the difference was significant (P=0.000). After purslane alkaloid intervention of breast cancer in nude mice, the pathology showed the reduction of tumor cells, accompanied by tumor necrosis and apoptosis. The results showed that the tumor tissue had high affinity for the purslane alkaloid. At the same time, it was also shown that the purslane alkaloid had a significant inhibitory effect on VEGF (P=0.000). Conclusion Purslane alkaloids can effectively inhibit the growth of breast cancer cells, and regulate the metabolism of tumor cells, which may be expected to become a new treatment methods and means of breast cancer.
