摘  要:目的:观察阿托伐他汀对沙鼠脑缺血再灌注损伤后脑组织中c-fos表达的影响及相应的保护机制。方法:健康雄性蒙古沙鼠制作脑缺血再灌注模型,缺血10min,再灌注6h、1d、3d、7d。采用HE染色法观察脑组织的病理变化,免疫组织化学染色法观察脑组织c-fos的表达情况。结果:(1)HE染色:脑缺血再灌注组根据时间点的不同可见胶质细胞增生、肥大,细胞间质和细胞水肿,神经元坏死。(2)免疫组化:缺血再灌注损伤后c-fos的表达上调,于第1天达到高峰,与对照组比较有显著差异,随后逐渐减少。阿托伐他汀干预组中c-fos表达趋势同缺血再灌注组,但各时间点阳性细胞数明显低于缺血再灌注组,相同时间点差异有显著性。结论:沙鼠前脑缺血再灌注后脑组织c-fos表达上调,阿托伐他汀下调c-fos表达,推测阿托伐他汀可能通过抑制c-fos的活性而起到凋亡负性调节的作用,从而减轻脑组织损伤程度。Objective:To observe the impact of atorvastatin on the expressions of c-los in the brain tissue following cerebral ischemic reperfusion in gerbils,and to study the neuroprotection mechanism of atorvastatin.Methods: Healthy male Mongolian gerbils underwent 10 minutes'common carotid artery occlusion(CAO) of both sides followed by reperfusion of 6 h, 1 d,3 d and 7 d respectively. The pathological changes of brain tissue and expressions of c-los were examined by HE staining and immunohistochemical methods. Results:(1) HE staining: The Ischemia Reperfusion Group were changed with gliocyte hyperplasia and hypertrophia, interstitial edema, cellular edema, gliocyte and neuron edema, neuron necrosis. (2)Immunohistochemistry:Compared with normal control group,the levels of c-los in neurons and gliacytes of ischemiwreperfusion gerbils were evidently upregulated at the 6th hour, 1st day, 3rd day and 7th day, and peaked at the 1st day, then downregulated gradually. Compared with I/R group,levels of c-fos in neurons and gliacytes of gerbils in atorvastatin intervention group were evidently decreased at the 6th hour, 1st day,3rd day and 7th day. Conclusion:Expressions of c-los in neurons and gliacytes of gerbils were sig- nifieantly increased following cerebral ischemia-reperfusion and injury of brain tissue after I/R in atorvastatin groups became moderate,which suggested that atorvastatin could have protected the brain tissue from damage through down-regulating the levels of c-los.
