摘  要:目的:建立决明子中葡萄糖、半乳糖、木糖、甘露糖、阿拉伯糖、葡萄糖醛酸、岩藻糖等7个单糖的HPLC测定方法,并比较其在生、炒决明子中的含量差别。方法:采用Kromasil C18色谱柱(150 mm×4.6 mm,5μm),流速为1.0 m L·min^(-1),检测波长为245 nm。流动相为乙腈(A)-水(B)、乙腈(A)-0.1%磷酸水(B),梯度洗脱,分别测定葡萄糖、半乳糖、木糖的含量与甘露糖、阿拉伯糖、葡萄糖醛酸、岩藻糖的含量。结果:生、炒决明子中葡萄糖、半乳糖、木糖、甘露糖、阿拉伯糖、葡萄糖醛酸、岩藻糖线性范围分别为0.028 8~1.15(r=0.999 6)、0.022 0~0.882(r=0.999 4)、0.025 8~1.03(r=0.999 5)、0.051 4~2.05(r=0.999 5)、0.010 6~0.426(r=0.999 3)、0.031 4~1.26(r=0.999 4)、0.047 8~1.19(r=0.999 3);加样回收率(n=5)在98.1%~99.3%,RSD在0.9%~1.7%。结论:炒决明子中单糖含量显著高于生决明子。Objective: To establish an HPLC method for determination of glucose,galactose,xylose,mannose,arabinose,glucuronic acid and fucose in Cassiae Semenand make a comparison between the crude and stirfrying Cassiae Semen. Methods: Chromatographic conditions concluded Kromasil C18( 150 mm × 4. 6 mm,5 μm) column at the flow rate of1. 0 m L/min. The detection wavelength was set at 245 nm. The mobile phaseⅠ was acetonitrile-0. 1% phosphoric acid in gradient elution mode for determination of glucose,galactose and xylose. The mobile phase Ⅱ was acetonitrile-water in gradient elution mode for determination of mannose,arabinose,glucuronic acid and fucose. Results: Glucose,galactose,xylose,mannose,arabinose,glucuronic acid and fucose had good linearity in the ranges of 0. 028 8 - 1. 15( r =0. 999 6),0. 022 0 - 0. 882( r = 0. 999 4),0. 025 8 - 1. 03( r = 0. 999 5),0. 051 4 - 2. 05( r = 0. 999 5),0. 010 6 - 0.426( r = 0. 999 3),0. 031 4 - 1. 26( r = 0. 999 4),0. 047 8 - 1. 19( r = 0. 999 3),respectively. The average recoveries of the seven constituents were 98. 1% - 99. 3% and RSDs were 0. 9%-1. 7%. Conclusion: The contents of seven monosaccharides in processed Cassiae Semen decreased.
