摘 要:目的:研究女贞子提取物(Ligustrum lucidum Ait Extract,LLAE)对链脲佐菌素(streptozocin,STZ)诱导的糖尿病肾病(DN)大鼠模型肾皮质血管内皮生成因子(VEGF)mRNA表达水平的影响,探讨女贞子提取物对肾脏的保护作用机制。方法:将65只SD大鼠随机分为正常对照组(N组),糖尿病肾病模型组(M组),女贞子提取物低(LLLAE,0.2 g/kg·d)、中(MLLAE,0.4 g/kg·d)、高剂量(HLLAE,0.8 g/kg·d)组,缬沙坦阳性对照组(V,10 mg/kg·d)。模型组、女贞子提取物低、中、高剂量组、缬沙坦组大鼠用链脲佐菌素(STZ)按55 mg·kg^(-1)空腹腹腔注射法构建糖尿病肾病大鼠模型。各实验组从造模成功后开始给药。实验第8周末处死大鼠,检测大鼠体重(BW)、肾脏肥大指数(KW.BW^(-1))、空腹血糖(FBG)、血尿素氮(Bun)、血清肌酐(Scr)和24 h尿白蛋白定量;HE染色法观察大鼠肾脏病理学变化;酶联免疫法和逆转录聚合酶链反应(RT-PCR)分别检测肾皮质VEGF蛋白含量和VEGFmRNA的表达。结果:女贞子提取物0.4、0.8 g·kg^(-1)剂量组与模型组相比体重明显上升,FBG、Bun、Scr、24 h尿蛋白定量、VEGF蛋白和VEGFmRNA表达显著降低,差别有非常显著意义(P<0.05或P<0.01)。结论:女贞子提取物可减少DN大鼠的尿蛋白排泄量、改善肾脏功能、延缓DN进程,其机制可能与通过降糖作用抑制了肾皮质内VEGFmRNA的表达水平,从而减少了VEGF的生成有关。Objective: To study the mechanism of extract of Ligustrum lucidum Ait in protecting the kidney by observing the effects of Ligustrum lucidum Ait extract on the expressions of VEGFmRNA in reanal cortical of rats with diabetic nephropathy. Methods: Sixty-five male rats were randomly divided into normal control group( N),diabetic nephropathy( DN) group,diabetic nephropathy group treated with Ligustrum lucidum Ait extract of low dose( 0. 2 g/kg),middle dose( 0. 4 g/kg) and high dose( 0. 8 g/kg). The diabetic nephropathy group was treated with valsartan( 10 mg/kg),ig,all once a day. DN group,three LLAE groups and valsartan group were intraperitoneal injected with STZ firstly. The rats were sacrificed after 8 weeks. The body weight,blood glucose,KW/BW,Bun,Scr and 24 h urinary protein were detected. The pathology changing of the rats' kidney was observed by HE staining. ELISA and RT-PCR were used to detect the expressions of VEGF protein and VEGFmRNA in reanal cortical of rats. Results: Compared with model group,the weight of LLAE 0. 4 g/kg and 0. 8 g/kg groups were increased significantly( P〈0. 05 or P〈0. 01). Compared with DN rats,LLAE 0. 4 g/kg and 0. 8 g/kg groups could reduce the levels of FBG,Bun,Scr and 24 h urinary protein and significantly suppress the over expression of VEGF in renal cortical tissue( P〈0. 05 or P〈0. 01). Conclusion: Ligustrum lucidum Ait extract can obviously reduce the urinary protein,improve the renal function and delay the progression of diabetic nephropathy in DN model rats. The kidney-protecting properties may be related to that LLAE can decrease the level of VEGFmRNA by reducing blood glucose level,and thus falling down the forming of vascular endothelial growth factor.
