葛根素对小鼠胚胎干细胞源心肌细胞超微结构的影响

摘  要:目的:探讨葛根素对小鼠胚胎干细胞源心肌细胞肌节及线粒体结构发育的影响。方法:采用悬滴-悬浮-贴壁三步法诱导转基因D3系小鼠胚胎干细胞向心肌细胞分化,在分化过程中持续使用葛根素(100μmol/L)处理;在分化的中期(第16天)和晚期(第20天)将拟胚体的跳动区域切下后制成超薄切片,应用电镜观察其超微结构,并测量肌节的长度。结果:⑴在分化第16天和第20天,葛根素组心肌细胞肌原纤维发育较对照组成熟;⑵分化第16天,葛根素组心肌细胞肌节长度为(1.67±0.02)μm,(n=9),明显长于对照组(1.44±0.15)μm,(n=4,P<0.05)。分化第20天,葛根素组心肌细胞肌节长度(1.88±0.04)μm,(n=6),高于对照组(1.78±0.04)μm,(n=5);⑶与对照组相比,葛根素组心肌细胞线粒体体积较大,嵴较密集。结论:葛根素能促进小鼠胚胎干细胞源心肌细胞肌节结构成熟和线粒体的发育。Objective To detect the effect of puerarin on sarcomere and mitochondria development of mouse embryonic stem cell-derived cardiomyocytes( m ESC-CMs). Methods The transgenic mouse embryonic stem cell line D3 was differentiated using the method of hanging drop-suspension-adherent,and the purerain( 100 μmol / L) was applied during the whole differentiation. The beating embryonic body areas were cut,prepared to ultrathin section,observed under electron microscope.The length of sarcomere was measured. Results ⑴At day 16 and day 20 of differentiation,the myofibril in the puerarin group was maturer than that in the control group; ⑵at day 16 of differentiation,the length of sarcomere in the puerarin group was( 1.67±0.02) μm,( n = 9),which significantly higher than that in control group( 1.44±0.15) μm,( n = 4,P〈0. 05). The length of sarcomere in the puerarin group( 1. 88 ± 0. 04) μm,( n = 6) also higher than that in control group( 1. 44±0.15) μm,( n= 4) at day 20 of differentiation; ⑶compared with control group,mitochondria were bigger and the mitochondria cristae were more intensive in puerarin group. Conclusion Our data showed that puerarin improves the ultrastructural development of m ESC-CMs.
