摘  要:目的:研究内皮祖细胞(EPC)移植对急性肺损伤(ALI)大鼠的抗炎效应。方法:将60只雄性SD大鼠随机分为对照组、ALI组、EPC组,每组各20只。ALI组和EPC组采用LPS(5 mg/kg)静脉注射制备ALI模型,EPC组在造模成功半小时后予EPC悬液移植,观察3组干预过程中血清TNF-α、IL-1β、IL-6和IL-10水平的变化,以及肺湿/干重比值(W/D)、TLR9表达和NF-κB活性的变化。结果:造模后ALI组和EPC组大鼠血清TNF-α、IL-1β、IL-6、和IL-10水平均显著升高(P<0.05)。造模7 d后,EPC组大鼠TLR9表达和NF-κB活性显著低于ALI组(P<0.05),血清TNF-α、IL-1β、IL-6水平也低于ALI组(P<0.05)。结论:EPC移植可以减轻ALI大鼠的炎症反应,具有一定的抗炎效应。Objective: To investigate the anti-inflammatory effect of endothelial progenitor cells (EPCs) transplantation in rats with acute lung injury. Methods: Sixty male SD rats were randomly divided into the control group, ALl group and EPC group (n:20 each).The ALl group and EPC group were treated with LPS (5 mg/kg) intravenously to prepare ALl model. The EPC group was given cell suspension transplantation at half an hour after successful modeling. The changes of serum TNF-α, IL-113, IL-6 and IL-10 levels, wet-to-dry lung weight (W/D) ratio, TLR9 expression and NF-KB activity during the intervention were determined in the three groups. Results: The serum TNF-α, IL-113, IL-6, and IL-10 levels in the ALl group and EPC group were significantly increased (P〈0.05). At 7 d after modeling, the expression of TLR9 and the activity of NF-KB in the EPC group were significantly lower than those in the ALI group (P〈0.05) , and the serumTNF-a, IL-113 and IL-6 levels inthe EPC group were also lower than those in the ALl group ( P〈0.05 ). Conclusion : EPCs transplantation can relieve inflammatory reaction in rats with acute lung injury with anti-inflammatory effect.
