摘 要:目的:观察坤泰胶囊对卵泡凋亡相关蛋白Bcl-2/Bax的影响。方法:建立卵巢早衰小鼠模型,造模成功后随机分为正常组、模型组、戊酸雌二醇片组、坤泰胶囊组。正常组、模型组给予生理盐水,戊酸雌二醇片组和坤泰胶囊组均以0.2 mL/10 g的体积灌胃给予相应药物。实验末,取卵巢、子宫称重,计算脏器指数,并以放免法和免疫组化法分别检测小鼠血清E2、FSH的水平和卵巢组织Bax、Bcl-2蛋白的表达。结果:坤泰胶囊组小鼠卵巢质量比模型组明显增加,卵巢指数与模型组相比差异具有显著性(P<0.05);坤泰胶囊组小鼠血清中E2水平比模型组小鼠明显升高、FSH水平明显降低,相对模型组差异具有显著性(P<0.05);坤泰胶囊组小鼠卵巢组织Bax蛋白表达比模型组明显降低,Bcl-2蛋白表达比模型组小鼠明显升高,与模型组比较差异均具有显著性(P<0.05)。结论:坤泰胶囊可以促进卵巢的生长,并通过调节卵泡凋亡相关蛋白Bcl-2、Bax的表达来抑制卵巢功能早衰。OBJECTIVE To explore the effects of Kuntai capsules on follicle apoptosis-related protein(Bcl-2/Bax)expression.METHODS Rats were divided into 4 groups:normal group,premature ovarian failure(POF)model group,Estradiol Valerate Tabletes group and Kuntai capsules group after inducing POF models successfully.Rats were given sodium chloride in normal group and POF model group,correspondent drugs in Estradiol Valerate Tabletes group and Kuntai Capsules group.After the experiment,ovarian index and womb index were studied.The levels of E2 and FSH were detected by RIA and levels of protein(Bcl-2/Bax)expression were detected by ELISA.RESULTS Compared with model group,Kuntai capsules group ovarian weight,and the ovarian index was increased(P〈0.05).The level of E2 was higher and level of FSH was lower in Kuntai capsules group(P〈0.05).The level of bax protein expression was significantly lower and Bcl-2 protein expression was significantly higher than that in model group(P〈0.05).CONCLUSION Kuntai capsules can promote the growth of ovarian,and decrease premature ovarian failure by regulating the expression levels of Bcl-2 and Bax.
