增加血浆含量对血小板保存质量的影响

摘　　要：目的探讨增加血浆含量对血小板体外保存质量的影响。方法将采集的12人双份共24个治疗量的血小板分为A、B 2组,A组为观察组血浆含量为280 mL/治疗量,B组为对照组血浆含量为200 mL/治疗量,2组均放置在(22±2)℃血小板保存箱震荡保存;分别在0、1、3、5 d时检测血小板计数、血小板平均体积(MPV)、pH值、血小板体外聚集反应和血小板活化率。结果 2组相比,血小板计数、MPV随时间的延长变化不显著(P>0.05);同一时间段A组的pH值较B组下降较慢,差异有显著性(P<0.05),血小板聚集反应随保存时间的延长而下降,A组较B组下降较慢(P<0.05),保存5 d时CD62p A组15.1±2.37(%),B组54.3±4.07(%),血小板活化率随保存时间的延长而升高,B组比A组升高较快(P<0.05)。结论增大保存血小板血浆的含量可以提高血小板体外保存质量。Objective To investigate the effect of increasing the content of plasma on the quality of platelet in vitro.Methods A total of 24 units of therapeutic dose platelets collected from twelve individuals were divided into two groups： A group was the observation group in which the plasma content was 280 m L per dosage; group B was control group in which the plasma content of 200 m L per dosage. Both two groups were placed in preservation at（ 22± 2） ℃. On days 0,1,3 and 5,platelet count,mean platelet volume（ MPV）,p H,in vitro platelet aggregation and platelet activation rate were obtained.Results Compared with the two groups,platelet count and MPV were not significantly changed（ P〈0. 05）. The B group was significantly lower than that in A group（ P〈0. 05）. The platelet aggregation reaction decreased with prolonged storage time（ P〈0. 05）. On day 5,the levels of CD62 p were 15. 1±2. 37（ %） and 54. 3±4. 07（ %） in the A and B groups,respectively. The platelet activation rate increased with prolongation of storage time,while the B group had significantly higher platelet activation rate than A group（ P〈0. 05）. Conclusion Increasing the plasma content in the preserved platelets can improve the quality of the platelet in vitro.
