摘 要:目的研究体外冲击波(extracorporeal shock wave,ESW)对大鼠软骨细胞的作用与影响。方法选取健康3周龄SD大鼠膝关节、股骨头软骨细胞进行原代培养,传至第2代时随机分为4组,待细胞贴壁1天后分别以能量为0、0.2、0.4、0.6 bar的ESW进行干预,干预后2天行CCK-8及Ⅱ型胶原蛋白蛋白质印迹法(Western blotting)检测。结果 CCK-8显示细胞活性逐渐增高,且有能量强度依赖性,在0.6 bar时作用最强(P<0.01)。0.2~0.6 bar能量的ESW同样可促进大鼠软骨细胞Ⅱ型胶原蛋白的表达,且该作用在0.6 bar时最为显著(P<0.01)。结论 0.2~0.6 bar能量的ESW可在一定程度上促进软骨细胞的增殖,增强软骨细胞Ⅱ型胶原蛋白的合成能力,且在0.6 bar时作用最显著。Objective To investigate the effect of extracorporeal shock wave (ESW) to the chondrocytes of rat. Method In the experiment, rat chondrocytes were divided into 4 groups. After one day, ESW with different energy (0, 0.2, 0.4, 0.6 bar) was given to each group. CCK-8 and Western blotting of type Ⅱ collagen were detected in two days later. Result The ESW energy from 0.2 to 0.6 bar could enhance the cell viability of chondrocytes, and had the dose-dependent,especially at the energy of 0.6 bar (P 〈 0.01). It could also promote the expression of type Ⅱ collagen, and the effect were remarkable when the energy were 0.6 bar (P 〈 0.01). Conclusion ESW energy from 0.2 to 0.6 bar can promote the cell viability of chondrocytes and enhance the expression of type Ⅱ collagen, especially at the energy of 0.6 bar.
