摘 要:目的探讨调脂通脉方对动脉粥样硬化兔模型血管重塑相关影响因素的调控机制。方法通过高脂饲料饮食及球囊损伤术建立动脉粥样硬化兔模型,54只兔随机分成5组,正常组、模型组、假手术组、阿托伐他汀组(2.2 mg/kg)和调脂通脉方组(8 g/kg);除正常组外,其余四组选择45天、90天两个观察点,45天时观察动物30只,90天时观察动物24只。对各组兔模型左颈总动脉的管腔内径、内中膜厚度和收缩期峰值流速采用超声测量;对颈总动脉作病理学观察;采用酶联免疫吸附测定法检测血清MMP-1、MMP-2、TIMP-1、PD-ECGF、UA水平。结果两个时间点,模型组兔血清中MMP-1、MMP-2、PD-ECGF、UA表达水平明显高于假手术组(P<0.01),TIMP-1低于假手术组(P<0.01)。调脂通脉方组家兔血清MMP-1、MMP-2、PD-ECGF、UA水平较模型组明显下降(P<0.01),TIMP-1水平明显升高(P<0.01),与阿托伐他汀组无明显差异。结论调脂通脉方降低了MMP-1、MMP-2、PD-ECGF、UA水平,提高了TIMP-1水平,从而影响动脉粥样硬化的血管重塑,进而改善动脉粥样硬化,可能是其作用机制。Objective To investigate the regulatory mechanism of Tiaozhi Tongmai formula on vascular remodeling related factors in rabbits with atherosclerosis. Method The rabbit model of atherosclerosis was established by high fat diet and balloon injury. 54 rabbits were randomly divided into normal group, model group, sham operation group, atorvastatin group and Chinese medicine group. Except the rabbits in normal group, the remaining four groups to choose the observation point at 45 day and 90 day after treatment. At 45 days, 30 animals were observed. At 90 days, 24 animals were observed. The lumen diameter and the intima-media thickness and the peak systolic velocity was measured by ultrasound. The common carotid arteries were observed by pathology. The levels of matrix metalloproteinases ( MMP-1, MMP-2) , tissue inhibitor of metalloproteinase-1 ( TIMP-1 ), platelet derived endothelial cell growth factor (PD-ECGF) and uric acid was detected by enzyme-linked immunosorbent assay(ELISA). Result There are two time points, t xpression levels of MMP-1 , MMP-2, PD-ECGF and UA in the model group were significantly higher than those in the sham operafio roup ( P〈0. 01 ), TIM P-1 was lower than that in sham operation group (P〈0.01). The serum levels of MMP-1, MMP-2, PD-ECGF and UA in the Tiaozhi Tongma formula group was significantly lower than those in the model group(P〈0.01 ), the level of TIMP-1 was significantly increased(P〈0.01 ), there was no significant difference from atorvastatin group. Conclusions Tiaozhi Tongmai formula can reduced the level of MMP-1, MMP-2, PD-ECGF, UA, and increase the level of TIMP-1, therefore influence the vascular remodeling to improve atherosclerosis, this may be the mechanism of its.
