摘  要:WRKY转录因子是植物界中存在的一类较大的基因家族,参与植物在生物和非生物逆境胁迫中的多种应答。为了研究在低温和高温等非生物胁迫对辣椒CaWRKY41基因表达的影响,以豫椒101为实验材料,在对辣椒CaWRKY41基因克隆的基础上,利用生物软件对其编码蛋白进行分析和构建进化树。结果表明,同源扩增获得的序列含有一个990 bp完整ORF框;对其编码蛋白分析发现,只含有一个WRKY结构域,属于WRKY41家族。亚细胞定位在细胞核中,分子进化树表明与茄属亲缘关系最近,相似性达到92%。荧光定量分析表明,与对照相比,低温、高温胁迫都能诱导CaWRKY41基因表达,表明CaWRKY41基因在应对低温和高温胁迫方面具有重要作用。WRKY transcription factor is a large gene family in the plant kingdom, which takes part in a variety of plant responses to the biotic and abiotic stress. In order to study the influence of abiotic stress such as low temperature and high temperature on the expression of CaWRKY41 gene in pepper, we used pepper 101 as experimental material to analyze the encoding protein and construct phylogenetic tree through bioinformatics software on the basis of the cloning of pepper CaWRKY41 gene. The results showed that the sequence obtained by homologous amplification contained a 990 bp complete ORF box; the analysis of the encoded protein revealed that it only contained one WRKY domain, which belonged to the family of WRKY4 l. Its subcellular localization was in the nucleus, and the molecular evolutionary tree showed that it was close to solanum, with the similarity of 92%. Fluorescence quantitative analysis showed that low and high temperature stress both could induce CaWRKY41 gene expression compared with the control group, which indicated that CaWRKY41 gene played an important role in response to low and high temperature stress.
