摘  要:目的:研究京尼平苷酸、齐墩果酸、补骨脂素、芝麻素、特女贞苷、肉桂酸6种补肾中药单体对氧化损伤内皮细胞中p66mRNA的调控,以探讨其抗氧化、保护血管内皮细胞的机制。方法:采用H_2O_2诱导EA.hy926(人脐静脉细胞融合细胞)为血管内皮细胞氧化应激损伤模型,予不同浓度的京尼平苷酸、齐墩果酸、补骨脂素、芝麻素、特女贞苷、肉桂酸干预后,用qPCR法检测不同单体对氧化损伤内皮细胞中p66mRNA的作用。结果:H_2O_2刺激EA.hy926后引起细胞内SOD活性降低,MDA含量增高,活性氧含量增高。5μM、50μM、500μM的京尼平苷酸、齐墩果酸、补骨脂素、芝麻素、特女贞苷、肉桂酸均能抑制血管内皮细胞氧化应激损伤模型中p66mRNA表达水平,其中以特女贞苷、齐墩果酸的作用效果最为明显。结论:补肾中药单体抗氧化、保护血管内皮细胞的作用与调节p66mRNA有关。Objective: To study the regulation of p66 mRNA of six kinds of tonifying kidney monomer: geniposidic acid, oleanolic acid, psoralen, sesamin, specnuezhenide, cinnamic acid on oxidative endothelial cells and to explore the mechanisms of its anti-oxidation and protection of vascular endothelial cells. Methods: EA. hy926 (human umbilical vein endothelial cells) were induced by H2O2 to be the oxidative stress damage model of vascular endothelial cells. After the intervention of different con- centrations of geniposidic acid, oleanolic acid, psoralen, sesamin, specnuezhenide, cinnamic acid, p66 mRNA in the oxidative endothelial cells was detected by qPCR assay. Results: The superoxide dismutase (SOD) activity was decreased. The content of malondialdehyde (MDA) and reactive oxygen species was increased after the stimulation of H2O2. 5μM, 50μM, 500μM of geni- posidic acid, psoralen, sesamin, cinnamic, especially acid specnuezhenide and oleanolic acid, inhibited the level of 1366 mRNA expression level of oxidative vascular endothelial cells model. Conclusions : The effect of anti-oxidation and protection of vascular endothelial cells of tonifying kidney monomer is related with the regulation of p66 mRNA expression level.
