摘 要:基因工程鲎素体外抑菌活性良好,为进一步评价其在体内抑菌活性和治疗效果,本研究建立了金黄色葡萄球菌小鼠肺部和腹腔感染模型,探讨了基因工程鲎素对2种感染模型小鼠的治疗效果。比较基因工程鲎素治疗前后,不同模型小鼠的存活率、血液以及组织中的细菌载量变化;比较治疗前后小鼠肺部感染动物模型的病理学变化。结果表明,与对照组相比,基因工程鲎素治疗后肺部感染模型小鼠存活率升高40%,腹腔感染模型小鼠存活率升高60%,治疗后腹腔感染小鼠血液中的细菌载量下降10~4倍;病理学分析结果表明,基因工程鲎素治疗后小鼠肺脏的炎症反应较轻,肺部组织结构被破坏程度较轻。综上所述基因工程鲎素对金黄色葡萄球菌感染小鼠的体内治疗效果明显,显著提高了小鼠存活率,抑制了细菌的扩散,减轻了病理炎症变化,初步表明基因工程鲎素具有良好的抗感染药物研发前景。Genetic engineering tachyplesin I has antibacterial activity in vitro, but the activity in vivo and therapeutic effect of bacterial infection are not clear. In this study, we constructed mouse Staphylococcus aureus pulmonary and abdominal infection models to explore the effect of genetic engineering tachyplesin I in vivo anti infection activity in mice. The survival rate of model mice, the bacterial load in the blood and tissue of different model mice were compared before and after treatment. The pathologic changes of the animal models of pulmonary infection before and after treatment were compared. The results showed that the survival rate of pulmonary model mice after treatment increased by 40% ,at the same time the survival rate of abdominal infection model mice after treatment increased by 60%; the levels of bacteria load in blood of abdominal infection model mice decreased significantly by 104 times. Lung HE staining sections of pulmonary model mice showed that the lung inflammatory response after treatment was mild, and the structure of lung tissue was not completely destroyed. In summary, curative effect of genetic engineering tachyplesin I against S. aureus in infected mice was remarkable, which can significantly improve the survival rate of mice, inhibit the spread of bacteria, reduce pathological changes. And preliminary results in- dicated that the genetic engineering tachyplesin I has a good prospect in anti infective drug research and development.
