摘  要:目的:探讨丁苯酞(dl-3-n-butylphthalide,NBP)对癫痫大鼠海马线粒体的保护作用。方法:(1)建立戊四氮(PTZ)点燃癫痫大鼠模型。(2)比色法测定海马线粒体呼吸链复合物Ⅱ,Ⅳ的活性。(3)透射电子显微镜观察海马CA3区线粒体形态。(4)BL-420生物机能系统采集并分析大鼠海马CA3区细胞电活动。结果:与癫痫组相比,丁苯酞组可显著提高癫痫大鼠海马线粒体呼吸链复合物Ⅳ的活性,差异有统计学意义(P<0.01);可减轻线粒体超微结构的损伤;并降低海马CA3区细胞群爆发性电活动的频率。结论:NBP对癫痫大鼠海马线粒体功能具有保护作用,从而减轻神经元异常电活动,可用于癫痫活动期的辅助治疗。Objective: To explore the protective effect of dl-3-n-butylphtalide( NBP) on hippocampal mitochondria in rats with epilepsy. Methods:(1) The kindling model was induced by pentylenetetrazol in rats.(2) The activity of mitochondrial respiratory chain complexes Ⅱ and Ⅳ in the hippocampus was determined by colorimetric method.(3) The mitochondrial ultrastructure of the hippocampal CA3 region was observed by transmission electron microscopy.(4) The electrical activity of hippocampal CA3 region was recorded using BL-420 Stereotaxic apparatus. Results: Compared with epileptic control,dl-3-n-butylphthalide significantly improved activity of the mitochondrial respiratory chain complex Ⅳ( P 〈0. 01),attenuated the damage degree of mitochondrial ultrastructure,and reduced the frequency of electrical explosive activity in hippocampal cells in CA3 region. Conclusion: Dl-3-nbutylphthalide has a protective effect on hippocampal mitochondrial function in epileptic rats,which can reduce the electrical activity of epilepsy in hippocampal CA3 region,and may be used as an adjuvant treatment of epilepsy.
