摘 要:目的探讨肺动脉高压患者肺组织1-磷酸鞘氨醇受体1/2/3(sphingosine-1-phosphate receptor1/2/3,S1PR1/2/3)的表达变化,为肺动脉高压发病机制的深入研究奠定基础。方法人的肺组织来源于黑龙江省医院,其中对照组取自右下肺叶球形性变实施肺叶切除患者的肺组织,病变组取自患有特发性肺动脉高压尸检患者的肺组织,应用免疫印迹及实时定量PCR技术检测S1PR1/2/3的表达变化。结果原发性肺动脉高压患者肺组织S1PR1/2/3蛋白的光密度值分别为2.00±0.03,0.86±0.13,0.94±0.03,m RNA的绝对定量值分别为3.90±0.42,2.50±0.37,92.08±11.26,较对照组蛋白的光密度值(0.88±0.29,0.27±0.07,0.54±0.06)及m RNA的绝对定量值(1.44±0.33,1.12±0.15,1.63±0.56)明显增高(P<0.01)。结论原发性肺动脉高压患者肺组织S1PR1/2/3的表达上调,提示S1PR1/2/3在肺动脉高压发病过程中可能发挥了重要的作用。Objective To investigate the expressions of sphingosine-1-phosphate receptor 1/2/3 in lung tissue of patients with pulmonary hypertension(PAH), to provide a foundation for further studying the pathogenesis of PAH. Methods The lung tissue was collected from Heilongjiang province Hospital, lungs of the control group were removed from patients with right lower lung lobe spherical lesions, and those of the PAH group from autopsy of patients with idiopathic primary PAH. Expression of S1PR1, S1PR2 and S1PR3 was evaluated using Western blot and real-time PCR. Results The optical density values of S1PR1, S1PR2 and S1PR3 proteins(2.00±0.03, 0.86±0.13, 0.94±0.03)and folder changes in m RNA(3.90±0.42,2.50±0.37, 92.08±11.26) in patients with primary PAH were significantly higher than those for proteins(0.88±0.29, 0.27±0.07, 0.54±0.06) and m RNAs(1.44±0.33, 1.12±0.15, 1.63±0.56) in the control group(P0.01). Conclusion The overexpressions of S1PR1, S1PR2 and S1PR3 in lung tissue of primary PAH indicate that they might play an important role in the pathogenesis of PAH.
