摘  要:目的探讨重组人促红细胞生成素(rHuEPO)对心梗大鼠心肌Fas与caspase-9表达的影响。方法 45只SD大鼠,随机分为假手术组、心肌梗死组和rHuEPO治疗组。用Western blot方法检测各组大鼠梗死区周围心肌中Fas与caspase-9的表达水平,RT-PCR方法检测梗死区周围心肌Fas mRNA与caspase-9 mRNA表达变化。结果心肌梗死组大鼠心肌梗死区周围心肌Fas与caspase-9蛋白与mRNA的表达水平显著高于假手术组,与心肌梗死组相比,rHuEPO治疗组心肌梗死区周围心肌Fas与caspase-9蛋白与mRNA的表达则明显降低(P<0.01)。结论 rHuEPO下调心梗大鼠心肌梗死区周围心肌Fas与caspase-9的表达。Objective To study the effect of recombinant human erythropoietin(rHuEPO) on the expressions of Fas and caspase-9 in myocardial infarction rats. Methods 45 SD rats were randomly divided into sham group, myocardial infarction group(MI group) and recombinant human erythropoietin group(rHuEPO group). The expressions of Fas and caspase-9 proteins and mRNA in tissue around infarction area were examined by Western blot and RT-PCR methods respectively. Result The expression levels of Fas and caspase-9 protein and mRNA in tissue around infarction area were increased significantly in MI group than in sham group, but decreased significantly in rHuEPO group than in MI group(P〈0.01). Conclusion rHuEPO could down-regulate the expressions of Fas and caspase-9 in myocardial infarction rats.
