摘 要:目的观察芪桂益脉灵对大鼠心肌缺血再灌注损伤基质金属蛋白酶9(MMP-9)和c-Jun氨基末端蛋白激酶(JNK)信号通路的影响。方法将50只健康雄性SD大鼠随机分为空白组(N)、假手术组(Sham)、单纯缺血再灌注组(I/R)、芪桂益脉灵高剂量组(QGYMLH)、芪桂益脉灵低剂量组(QGYML),每组10只,分别给予0.9%氯化钠注射液、芪桂益脉灵(高、低剂量),连续灌服7 d,结扎冠状动脉左前降支造成急性心肌梗死模型,腹主动脉采血,离心取上层血清,采用酶联免疫法检测MMP-9的表达;取心肌组织,用中性福尔马林固定,应用免疫组化法检测磷酸化c-Jun氨基末端蛋白激酶(P-JNK)表达。结果 Sham组MMP-9、P-JNK表达水平与N组比较差异无统计学意义(P>0.05);I/R组、QGYMLL组、QGYMLH组MMP-9、P-JNK表达水平均高于N组(P<0.05);QGYML组、QGYMLH组MMP-9、P-JNK表达水平均低于I/R组(P<0.05);QGYMLH组MMP-9、P-JNK表达水平低于QGYMLL组(P<0.05)。结论芪桂益脉灵对缺血再灌注损伤心肌具有保护作用,其机制与抑制心肌组织中JNK磷酸化程度及MMP-9表达有关,且与中药剂量相关。Objective To observe the effects of Qigui-yimailing on the signal pathway of matrix metalloproteinase-9(MMP-9) and c-Jun amino terminal protein kinase(JNK) in rats with myocardial ischemia reperfusion injury. Methods 50 healthy male SD rats were randomly divided into normal group(N),sham-operated group(Sham),pure ischemia reperfusion group(I/R),high dose of Qigui-yimailing group(QGYMLH) and low dose of Qigui-yimailing group(QGYMLL),10 rats each group,rats in last three group were received 0. 9% sodium chloride injection,high and low dose of Qigui-yimailing respectively,continuously treatment for 7 d. The acute myocardial infarction model was established by ligating the left anterior descending coronary arteries. The blood was collected from the abdominal aorta,the upper serum was taken by centrifugal,and the expression of MMP-9 was measured by ELISA. The expression of phosphorylation c-Jun amino terminal protein kinase(P-JNK) from formalin-fixed myocardial tissue was measured by means of immunohistochemical staining. Results There were no statistical differences on the expressions of MMP-9 and P-JNK between Sham and N group(P〈0. 05). The expressions of MMP-9 and P-JNK in I/R,QGYMLL and QGYMLH group were higher than N group(P〈0. 05). The expressions of MMP-9 and P-JNK in QGYMLL and QGYMLH group were lower than I/R group(P〈0. 05). The expressions of MMP-9 and P-JNK in QGYMLH group were lower than QGYMLL group(P〈0. 05). Conclusion Qigui-yimailing has protective effect of myocardial ischemia-reperfusion injury,and its mechanism may be related with the degree of JNK phosphorylation,the MMP-9 expression and the dosage of drug.
