补肾健脾利湿化瘀法对IgA肾病模型大鼠血清MMP-9、TIMP-1含量的影响

摘　　要：目的探讨补肾健脾、利湿化瘀法对Ig A肾病模型大鼠血清MMP-9、TIMP-1含量的影响。方法将80只Wistar大鼠随机分为空白组10只,造模组70只,造模成功后随机分为模型组,血尿胶囊对照组,补肾健脾、利湿化瘀法中药汤剂高、中、低剂量组(简称中药高、中、低剂量组)。检测大鼠各时相尿红细胞计数,血清中MMP-9、TIMP-1含量及24 h尿蛋白定量。结果 1)空白组大鼠一般状态良好,造模后大鼠第4周开始出现精神不振,进食及活动减少,毛色晦暗,尿色改变,体质量下降。给予药物治疗后,各症状均有所改善;2)与空白组相比较,造模后大鼠尿红细胞计数及24 h尿蛋白定量自第6周开始升高,至第8周达到高峰;与模型组比较,各组大鼠尿红细胞计数降低(P<0.05),组间比较,中药中剂量组明显优于血尿胶囊组。3)与空白组比较,造模后各组大鼠血清MMP-9含量降低,TIMP-1含量升高,MMP-9/TIMP-1降低(P<0.05);与模型组相比较,各治疗组大鼠血清MMP-9含量明显升高,TIMP-1含量明显降低,MMP-9/TIMP-1升高(P<0.05),组间比较,中药中剂量组明显优于血尿胶囊组(P<0.05)。结论 1)补肾健脾、利湿化瘀法能够降低Ig A肾病模型大鼠尿红细胞计数及24 h尿蛋白定量;2)造模组大鼠血清中MMP-9含量降低,TIMP-1含量升高,MMP-9/TIMP-1降低(P<0.05),说明MMP-9、TIMP-1参与了Ig A肾病发病机制;3)给予药物治疗后,各治疗组大鼠血清MMP-9含量明显升高,TIMP-1含量明显降低,MMP-9/TIMP-1升高(P<0.05),组间比较,中药中剂量组疗效最为显著,明显优于血尿胶囊组。笔者推测补肾健脾,利湿化瘀法治疗Ig A肾病的作用机制可能是调整血清中MMP-9、TIMP-1的含量。Objective To study the influence of Bushenjianpi and Lishihuayu methods to serum levels of MMP-9 and TIMP-1 content of IgA nephropathy model rats. Methods 80 Wistar rats were randomly divided into blank group( n =10) and build module(n =70). After modeling, the rats were randomly divided into model group and hematuria capsule control group, Bushen Jianpi and Lishi Huayu methods high, medium and low dose group(which were called traditional Chinese medicine high, medium and low dose group). The red blood cell count, serum MMP-9, TIMP-1 and 24 h urine protein were measured in each phase on rats. Results 1) The rats in the blank group were in good condition. After 4 weeks of modeling, the rats began to be mentally depressed, eating and activity decreased, the coat color was dull, the urine color was changed and the body weight was decreased. After the drug treatment, the symptoms improved. 2) Compared with the blank group, the urinary red blood cell count and 24 h urine protein in the rats were increased from the 6th week to the 8th week; Compared with the model group, the erythrocyte count of the rats in each group decreased(P <0.05), the comparison between the groups, the traditional Chinese medicine dose group was superior to the hematuria capsule group. 3) Compared with the blank group, after building groups of rats serum content of MMP-9 is reduced, TIMP-1 content increases, MMP-9/TIMP-1 lower(P <0.05); Compared with model group,the treatment group rats serum content of MMP-9 increased significantly, content of TIMP-1 reduced significantly, MMP-9/TIMP-1 elevated(P<0.05), the comparison between groups, traditional Chinese medicine in the dose group is superior to hematuria capsule group(P <0.05). Conclusion 1) Bushen Jianpi and Lishi Huayu methods can reduce the urine red blood cell count and 24 hours urine protein quantitative of IgA nephropathy model in rats; 2) In build module, MMP-9 content in serum of rats decreased, TIMP-1 content increases, MMP-9/TIMP-1 lower(P < 0.05), MMP-9 and TIMP-1 is involved in the
