摘 要:目的研究霉茶总黄酮提取物对自发性高血压大鼠(SHR)胸主动脉重构的影响及其机制。方法将SHR随机分为模型组,总黄酮提取物低剂量组(90 mg·kg-1)、中剂量组(180 mg·kg-1)、高剂量组(360 mg·kg-1)和复方罗布麻片组(50 mg·kg-1)。每日给药2次,连续7周,末次给药前禁食12h,末次给药后1 h,处死大鼠,取胸主动脉。部分胸主动脉做病理组织学检查,部分胸主动脉测定其组织ACE、AngⅡ、C-myc、Bcl-2、P27KIP1基因的表达。结果霉茶总黄酮提取物中、高剂量能明显降低血管中膜厚度与管腔内径比值,霉茶总黄酮提取物能下调血管组织中ACE、AngⅡ、C-myc、Bcl-2m RNA表达,同时上调P27KIP1基因表达。结论霉茶总黄酮提取物能抑制SHR的血管重构,其机制与调节RAS系统相关基因及VSMCs增殖相关基因有关。Objective To determine the effect and mechanism of total flavonoids extract of ampelopsis megalophylla on thoracic aortic reconstruction in spontaneously hypertensive rats(SHR). Methods The SHRs were randomly divided into a model group, a total flavonoids extract low-dose group(90 mg·kg^-1), a total flavonoids extract medium-dose group(180 mg·kg^-1), a total flavonoids extract high-dose group(360 mg·kg^-1), and a Fufang Luobuma tablet group(50 mg·kg^-1). Drugs were administered orally twice daily for 7 weeks. The rats were fasted 12 h before the last administration. One hour after the last administration, the rats were sacrificed and the thoracic aorta were taken. Morphological indexes of the thoracic aorta were measured. ACE, AngⅡ, C-myc, Bcl-2, and P27KIP1 gene expression in the thoracic aorta were also measured. Results Total flavonoids of ampelopsis megalophylla extract significantly reduced vascular thickness and lumen diameter ratio in both high-dose and medium-dose groups. And total flavonoids of ampelopsis megalophylla extract reduced ACE, AngⅡ, C-myc and Bcl-2 gene expression, and also raised P27KIP1 gene expression. Conclusion Total flavonoids of ampelopsis megalophylla extract can inhibit vascular remodeling in SHRs, which is associated with regulation of RAS system-related genes and VSMCs proliferation related genes.
