摘 要:目的探讨甘草酸铵对对乙酰氨基酚(APAP)诱导人正常肝细胞损伤的保护作用。方法用不同浓度的APAP来制备人肝细胞损伤模型,不同浓度的甘草酸铵处理,噻唑蓝(MTT)法检测人肝细胞的存活率,ELISA法检测细胞培养上清中谷丙转氨酶(ALT)和天门冬氨酸氨基转移酶(AST)的含量。结果与正常组比较,用不同浓度的APAP处理的人肝细胞存活能力显著降低,且APAP致人肝细胞损伤的最佳造模条件为15 mmol·L-1孵育12 h。与模型组比较,给予不同浓度的甘草酸铵能显著提高APAP损伤细胞的存活率,改善受损肝细胞的形态,降低ALT和AST的释放量。结论甘草酸铵对APAP损伤的肝细胞具有很好的保护作用,为甘草酸铵治疗药物性肝损伤提供依据。Objective To investigate the protective effect of ammonium glycyrrhizinate(AG) on injured human hepatocytes induced by acetaminophen(APAP). Methods The injured hepatocytes model was established by different concentrations of APAP. The injured hepatocytes were treated by AG. The cell viability was detected by MTT assay, and concentrations of ALT and AST were assessed by ELISA. Results Compared with the control group, the cell survival of injured human hepatocytes induced by different concentrations of APAP was significatly reduced, and the most optimum condition was 15 mmol·L^-1, culturing for 12 h. Compared with the model group, AG promoted the cell viability, improved the cells morphology, decreased the concentration of ALT and AST. Conclusion AG exerts protective effect on injuried human hepatocytes induced by APAP, which provides evidences for treatment of drug-induced liver injury.
