摘  要:目的:检测龟甲胶中新阿胶特征肽成分。方法:采用胰蛋白酶进行酶解,利用超高效液相色谱-电喷雾四极杆飞行时间质谱法(UPLC-ESI-QTOF-MS/MS)对龟甲胶样品进行测定,选定新阿胶特征肽离子m/z925.4作为检测离子。结果:龟甲胶特征成分含量偏低的样品(但不含有牛皮元和驴皮元特征肽成分)中检测出新阿胶特征肽离子m/z 925.4信号,其MS/MS裂解规律与文献报道一致,而龟甲胶标准品中未检测出该特征肽信号。结论:该方法专属性强、灵敏度高,可检测出龟甲胶中是否掺杂新阿胶成分,可用于龟甲胶的质量控制。This study aimed at identifying pig-hide gelatin from tortoise shell glue using ultra-performance liquidchromatography-electrospray-time of flight mass spectrometry(UPLC-ESI-QTOF-MS/MS).The samples of tortoise shellglue were digested by trypsin before the analysis on UPLC coupled with QTOF mass spectrometry. The ions at m/z 925.4were selected as the marker peptides of pig-hide gelatin. It was found that the test results of the ions at m/z 925.4 werepositive in the samples with low contents of the marker peptides of tortoise shell glue, indicating the existence of pig-hidegelatin. However,the result of bovine-hide gelatinordonkey-hide gelatin was negative, while negative results weredetected in tortoiseglue standard. The finding was in conformity with previous references over the fragmentation regularityof MS/MS. In conclusion, the method can be adopted to identifying the pig-hide gelatin from tortoise shell glue andapplied to the quality control of tortoise shell glue with high specificity and sensibility.
