摘 要:应用RT-PCR方法对分离自广西不同鸽场的8株鸽源NDV广西分离株HN基因进行扩增、序列测定和分析,旨在探讨广西鸽源NDV HN基因的分子进化特点,为科学防控鸽源新城疫提供依据。结果显示,8株鸽源NDV广西分离株HN基因的ORF全长均为1 716 bp,编码571个氨基酸,属于强毒的C群,符合强毒株的基因长度特征。核苷酸同源性比较显示,8株鸽源NDV分离株与NDV基因Ⅵ型中的Ⅵb亚型同源性最高,为90.4%~99.5%。遗传进化分析显示,8株鸽源NDV广西分离毒株与我国2011-2013年广西、广东、吉林、辽宁、云南、黑龙江等地鸽源NDV分离株的遗传关系较为接近,位于同一进化树分支上,初步推测8株鸽源NDV广西分离株应属于classⅡ基因Ⅵb亚型NDV。The HN gene of eight NDV strains iso|ated from Guangxi different pigeon farms, were amplified by RT -PCR metbod, sequenced and analyzed. The aim of this study is to explore the molecular evolution characteristics of Guangxi pigeon NDV HN gene, and to provide the basis for scientific prevention and control of Pigeon Newcastle disease. The results showed that the nucleotide sequence length of HN geue of the eight NDV isolates was 1716 hp,encoding 571 amino acids. They belonged to the virulent C group, and the gene length characteristic of HN gene of the eight NDV isolates was in accord with the virulent strain. The analysis of nucleotide homologies indicated that the eight NDV isolates shared higher homology with genotype Vlb,ranging from 90. 4% to 99. 5%. The phylogenetie tree analysis demonstrated that the genetic relationship between the eight guangxi NDV strains and the isolates from Guangxi,Guangdong,Jilin, Liaoning, Yunnan, and Heihmgjiang from 2011 to 2013 was close. They were located in the same cladogram branch. We assume that the eight pigeon NDV isolates isolated in Guangxi all belong to the gene class Ⅱ genolype Ⅵb NDV.
