摘 要:丛枝菌根真菌(Arbuscular mycorrhizal fungi,AMF)通过参与植物碳氮代谢及其渗透调节物质影响植物的抗逆性。为进一步评价AMF玫瑰红巨孢球囊霉(Gigaspora rosea)调控烟草植物的抗逆性提供依据,本试验设计用孔径为30μm的尼龙网分隔的分室系统,将烟草(Nicotiana tabacum L.)定植于分室系统中的菌根室,陷阱室设置^(15)N同位素陷阱。菌根室接种100 g AMF Gigaspora rosea菌剂(其中含有活性孢子约100个,菌根根段和菌丝),对照接种相同剂量的灭菌菌剂。培养3个月后检测烟草的营养生长、氮代谢关键酶活性、稳定同位素^(15)N、总氮、渗透调节物质及叶绿素含量。结果表明,与对照相比,接种AMF的烟草,菌根侵染率高达69.12%,显著促进了烟草生长;根、茎、叶中都检测到了稳定同位素^(15)N,^(15)N丰度大小及相对偏差顺序为叶>根>茎;接种AMF增强了烟草植株氮代谢关键酶活性,总氮、精氨酸、总叶绿素、可溶性蛋白、脯氨酸含量分别增加了31.74%、34.71%、32.11%、47.56%和20.13%,烟草中精氨酸含量与总氮水平正相关,接种下R^2≥0.932 5。结果证明AMF通过促进氮代谢及提高烟草渗透调节物质含量来提高烟草的抗逆能力。Arbuscular mycorrhizal fungi (AMF) affected plant resistance through taking part in plant carbon and nitrogen metabolism and adjustment of osmotic substances. In order to provide the basis for further evaluation of Gigaspora rosea on regulating and controlling resistance of tobaccos, the special compartment container was designed to be separated evenly into mycorrhizal compart- ment and 15N trap compartment by 30 μm nylon mesh. Two tobacco seedlings were planted in my- corrhizal compartment which contained sterile matrix and inoculated or not with 100 g inoculum of arbuscular mycorrhizal fungi Gigaspora rosea. 15N traps were lain up on the bottom of trap compart- ment. After 3 months of routine culture, the vegetative growth, the key enzyme activity of nitrogen metabolism (Glutamate synthase and Glutamine synthetase), the contents of stable isotope, total nitrogen, arginine, osmotic adjustment substances, and chlorophyll of tobaccos were tested. The results showed that the mycorrhizal infection rate of tobaccos inoculated with AMF was as high as 69.12%, which significantly promoted the vegetative growth of tobaccos. The stable isotope 15N were detected only in the roots, stems and leaves of tobaccos. The distribution of the 15N abundance was leaf 〉 stem 〉 root. The key enzyme activity of nitrogen metabolism significantly was promoted because the content of total nitrogen, arginine, chlorophyll, soluble proteins and proline respectively were increased by 31.74%, 34.71%, 32.11%, 47.56% and 20.13% in inoculated to- baccos compared to the uninoculated one with Gigaspora rosea. Meanwhile, the content of arginine was positively related to the total nitrogen in tobaccos, under inoculated conditions, R2 ≥0.932 5. The results confirmed that AMF contributed to the resistance ability of tobaccos by promoting the nitrogen metabolism and improving the contents of osmotic adjustment substances.
