摘  要:目的探讨温浴不同温度和时间对血浆中白细胞介素8(interleukin-8,IL-8)的影响,为库存红细胞安全、有效的使用提供理论依据。方法随机抽取河北省血液中心采集的无偿献血、经检测各项指标均合格的血液标本50份,每份均分成8段(A、B、C、D、E、F、G、H组)分别行4℃保存,37℃加热10、20、30、60、120、180min和40℃加热30min处理,并进行后续实验。采用双抗体夹心酶联免疫吸附测定法检测血浆中IL-8浓度。结果加热能明显上调IL-8水平,并且有时间依赖性,随加热时间延长IL-8水平逐渐增高,B、C组血浆IL-8浓度与A组相比差异无统计学意义(P>0.05),而D、E、F、G、H组血浆IL-8浓度与A组相比显著升高,差异有统计学意义(P<0.05)。结论库存红细胞37℃温浴30min以内对血浆IL-8浓度无明显影响,之后随着时间延长逐渐升高。Objective To study the effects of storing suspending erythrocytes after water bath on interleukin-8(IL-8)in plasma,and further more to confirm the effect of water bath at various times and different degrees on IL-8in plasma.Our study will provide the theoretical basis for safe and effective use of storing suspending erythrocytes.Methods Fifty blood samples were selected randomly from blood center in HebeiProvince,with detection of all indicators qualified.Each divided into 8groups(A,B,C,D,E,F,G,H)which were stored at 4 ℃,heated at 37 ℃for various time(10,20,30,60,120,180 min)and 40 ℃ for 30 min for subsequent experiments.IL-8concentration in plasma was detected by adopt double antibody sandwich enzyme-linked immunosorbent assay.Results Heating could significantly up-regulated IL-8level,and with time-dependence pattern.There was no significant difference in plasma IL-8concentration between B and C group compared with A group(P>0.05).Plasma levels of IL-8in D,E,F,G,H were significantly higher than those in the A group,the difference was statistically significant(P<0.05).Conclusion There was no significant effect on plasma IL-8concentration in water bath at 37 ℃ within 30 min,and then gradually increased with time extended.
