摘  要:目的研究黄芩提取液体外抑菌的最佳提取工艺及黄芩苷的细胞保护作用。方法采用正交实验设计,以黄芩提取液的最低抑菌浓度(MIC)及黄芩苷的含量为指标,筛选最佳提取工艺。以MTT法测定黄芩苷对TNFα/INFγ联合刺激人皮肤角质细胞(HACAT)的保护作用。结果综合极差数据、抑菌实验结果,确定工艺路线为A_3B_3C_2,即:10倍量60%乙醇,提取2次,每次1.5 h。黄芩提取液对金黄色葡萄球菌和表皮葡萄球菌均有良好抑制作用,黄芩苷对TNFα/INFγ刺激HACAT细胞引起的炎症反应具有明显的抑制作用。结论提取方法稳定合理,提取液具有良好的抗菌作用,黄芩苷也有较好的抗炎作用。OBJECTIVE To study the optimal extraction process of Scutellaria baicalensis and the functional effect of baicalin in vitro. METHODS The extract solution of Scutellaria' s MIC and Baicalin' s content were used to screen the optimal extraction process with Orthogonal experimental design. HACAT cells were induced by TNFα/INFγ, and baicalin' s cytoprotection was studied by MTT. RESULTS The optimal extraction solution of S. baicalensis could inhibit Staphylococcus epidermidis and Staphylococcus aureus efficiently. Baicalin could reduce the TNFcα/INFγ mediated damages to HACAT cells. CONCLUSION This extraction process is rea- sonable and stable, and the optimal extraction solution has good bacteriostasis. Baicalin has cytoprotection on the HACAT ceils induced by TNFα/INFγ.
