摘 要:目的观察电针内关预处理对大鼠缺血再灌注损伤心肌的影响,并探讨电针预处理防治心肌缺血再灌注损伤的作用机制。方法将40只SD雄性大鼠随机分为假手术组、缺血再灌注组(模型组)、电针内关穴组和电针环跳穴组,每组10只。采用冠脉结扎法造模,电针内关组和电针环跳组在造模前,分别给予电针刺激每日20 min,共7 d。记录造模前后心电图Ⅱ导联T波电压值,透射电镜检测心肌病理形态学的变化,采用硝酸还原酶化学比色法检测血清一氧化氮(NO)、一氧化氮合酶(NOS)的含量,免疫组化法检测心肌组织腺苷A1受体含量。结果模型组血清NO含量、NOS活性及腺苷A1受体较假手术组明显下降(P<0.05或P<0.01);电针内关穴组血清NO含量、NOS活性及腺苷A1受体较模型组和电针环跳组明显升高(P<0.01或P<0.05)。模型组大鼠与电针环跳穴组大鼠比较差异无统计学意义(P>0.05)。结论电针内关穴可以有效改善心肌缺血再灌注损伤,对心肌产生保护作用。Objective: To explore the cardioprotective mechanisms of electroacupuncture(EA) at Neiguan point and observe the effect of it on NO,NOS and adenosine A1 receptors in rats with myocardial ischemia-reperfusion injury(MIRI). Methods: 40 SD male rats were randomly divided into the sham group,model group,EA Neiguan group and EA Huantiao group,10 rats in each group. The models were established by ligating the coronary artery,The EA Neiguan group and EA Huantiao group were given electroacupuncture stimulation pretreatment 20 min every day lasting for 7 days before establishing MIRI model respectively. II T wave of lead electrocardiogram(ecg)before and after model was recorded,transmission electron microscopy(sem) was used to detect myocardial pathomorphism changes. The content of serum NO and NOS was determined by ELISA method,Immunohistochemical method was used to detect the contents of myocardial tissue adenosine A1 receptors. Results: NO,NOS and adenosine A1 receptors of the model group decreased significantly than those of control sham-operation group with obviously statistical difference(P〈0.05);NO,NOS contents and adenosine A1 receptors of EA Neiguan group increased significantly than those of model group,control sham-operation group and EA Huantiao group with statistical difference(P〈0.01 or P〈0.05);but there was no significant difference between model group and EA Huantiao group(P〈0.05). Conclusion: The EA at Neiguan point can improve the MIRI and shows cardioprotective effects.
