摘　　要：通过研究1型糖尿病(T1DM)猕猴肝内大麻素1型受体(CB1R)、脂肪酸酰胺水解酶(FAAH)和糖皮质激素受体(GR)蛋白和mRNA表达量的变化,探讨内源性大麻素系统对糖尿病猕猴肝的影响。苏木精-伊红染色观察糖尿病猕猴肝的病理变化,均可见肝细胞肿胀伴气球样变,不同程度脂肪变性。油红O染色结果显示,糖尿病猕猴组猕猴肝中脂滴含量显著高于对照组(P<0.01)。免疫组织化学结果显示,糖尿病组猕猴肝中CB1R表达量高于对照组(P<0.05),而糖尿病组猕猴肝中FAAH、GR表达量均低于对照组(P<0.05)。实时荧光定量PCR结果显示,CB1R、GR、FAAH的mRNA的表达量变化与蛋白表达一致,对照组与糖尿病组差异相比有高度统计学意义(P<0.01)。结果表明,CB1R、GR、FAAH与T1DM猕猴肝病理损伤密切相关,可为临床上该疾病的监测与治疗提供依据。In order to investigate the relationship between endocannabinoid system and the liver of type 1 diabetes mellitus（T1DM）macaques,we designed to test the levels of three proteins and their m RNA,including cannabinoid type 1 receptor（CB1R）,fatty acid amide hydrolase（FAAH）and glucocorticoid receptor（GR）in the liver of T1DM macaques.It was observed that the hepatocellular swelling,ballooning and steatosis appeared in the liver of T1DM macaques by HE staining.Oil red O staining showed that the lipid droplets in the liver T1DM group were significantly higher than those in the control group（P〈0.01）.Immunohistochemistry demonstrated that the expression of CB1 R in the liver of T1DM group was higher compared with the control group（P〈0.05）,but the expression of FAAH and GR in liver of diabetic group was lower on the contrary（P〈0.05）.Real-time fluorescent quantitative PCR results suggested that the expression of CB1 R,GR and FAAH m RNA was similar to the protein expression,and the difference between the control group and the T1DM group was extremely significant（P〈0.01）.All the results showed that CB1 R,GR and FAAH were closely correlated to liver injury in T1DM macaques,and could provide basis for the clinical monitoring and treatment of the disease.
