摘 要:目的比较丙泊酚和水合氯醛对亚低温大鼠神经细胞凋亡的干预作用,促进丙泊酚在临床低温麻醉手术中的合理应用。方法选择健康雄性SD大鼠30只,随机分为水合氯醛常温组(A组)、丙泊酚低温组(B组)、水合氯醛低温组(C组),各10只。A组保持正常体温,B组和C两组低温干预30 min,各组进行心脏灌注,断头取脑,制备大鼠海马神经元组织样本,分别进行凋亡标志物Caspase-3检测及透射电镜超微结构观察,对比丙泊酚与水合氯醛对亚低温所致大鼠海马神经元凋亡的影响。结果用平均光密度(AOD)值半定量Caspase-3蛋白的表达,结果水合氯醛低温组表达明显低于其常温组,但高于丙泊酚低温组(P<0.01);透射电镜结果显示,水合氯醛常温组海马区神经元有明显的凋亡小体形成,而水合氯醛低温组也有不同程度的凋亡趋势,只有丙泊酚低温组凋亡现象最轻。结论大鼠亚低温状态海马神经元仍存在一定的凋亡现象,而丙泊酚可减少亚低温状态下海马神经元的凋亡,同等条件下水合氯醛对海马神经元的凋亡则无明显的干预作用。Objective To compare the intervention effect of Propofol and Chloral Hydrate on the apoptosis of nerve cells in mild hy- pothermia rats, in order to promote Propofol better applied in clinical hypothermic anesthesia surgery. Methods Totally 30 health SD rats were selected and randomly divided into normal temperature group of chloral hydrate(group A), low temperature group of propofol group(group B) and low temperature group of chloral hydrate(group C),10 cases in each group. The body temperature of group A was maintained normal,and those of group B and group C were under low- temperature intervention for 30 min,each group underwent heart perfusion, the brains were taken after decapitation to prepare the rat hippocampal neuronal tissue samples, and then apoptosis marker Caspase- 3 was detected and the ultra- microstructure was observed by transmission electron microscopy to compare the influence of Propofol and Chloral Hydrate on the apoptosis of nerve cells in mild hypothermia rats. Results The semi- quantification was carried out on the expressions of Caspase-3 protein by average optical density(AOD). The result showed that the expression of Caspase-3 protein in group C was significantly lower than that in group A,but higher than that in group B (P 〈 0. 01);The results of the trans- mission electron microscopy showed that there were obvious apoptotic bodies formed in the hippocampal neurons in group A,while there were also different degrees of apoptotic tendency in group C, and the apoptosis phenomenon was only not serious in group B. Conclusion There are certain of apoptosis phenomena in the hippocampal neurons in hypothermia rats,while the Propofol can reduce the apoptosis of hippocampal neurons under hypochromic condition, and the Chloral Hydrate has no obvious intervention effect on the apoptosis of hippocampal neurons at the same condition.
