摘 要:目的:以五味子为原料,以总木脂素为研究对象,展开了对总木脂素的提取、分离纯化及还原能力的研究。方法:本文通过静态和动态实验对5种大孔树脂进行筛选,以吸附、解析数据为指标,优选大孔树脂纯化工艺,并利用Fe3+法测定总木脂素体外还原能力。结果:确定AB-8型树脂最优,最佳工艺条件为:五味子总提取物上样浓度为1.5 mg/m L,流速为1 BV/h;最终用50%乙醇(低浓度洗脱剂)和70%乙醇(高浓度洗脱剂)进行洗脱;体外还原能力结果显示:五味子总木脂素在50%和70%乙醇组分对Fe3+表现出较高的还原能力。试验结果显示,通过大孔树脂纯化五味子总木脂素及其体外还原能力的研究,五味子总木脂素的含量从11.68%提高到41.48%。结论:本实验的梯度洗脱法可显著提高五味子中总木脂素的质量分数,并且五味子总木脂素在50%和70%乙醇组分表现出良好的体外还原能力,为今后五味子总木脂素研究提供理论依据。Objective:It is Schisandra chinensis as raw materials and the total lignans for the study that we research the total lignans element of extraction,isolation,purification and reducing power method. Methods:Taking Schizandrin as test index that used the UV spectrophotometry method for measuring the total lignans content from Schisandra chinensis(Turcz);The static purification condition of lignans were determined on the basis of investigating the adsorption and desorption performance of different kind of macroporous resins. Using Fe3+ method research the total lignans reducing ability in vitro. Results:AB-8 macroporous resin was chose and employed in this study. The optimal adsorption conditions were as follows:in order to obtain lignin with the density of 1.5 mg/m L. Best elution method is 1 BV/h flow rate in turn with 50% ethanol and 70% ethanol elution. In vitro reducing power results showed that Schisandra lignans showed higher reducing ability in 50% ethanol and 70% ethanol. The test results show that the lignans content of Schisandra crude extracts increased from 11.68% to 41.48%. Conclusion:The method of gradient elution is a better way to purify total lignans Schisandra chinensis(Turcz.)Baill. The total lignans from Schisandra chinensis(Turcz.)Baill in 50% and 70% ethanol component showed good in vitro reduction ability,which provided a theoretical basis on total lignans of Schisandra chinensis(Turcz.)Baill for the future study.
